Objective:To analysis the differences of protein expression of renal tumor tissue, cancer-adjacent and normal tissue and to confirm the optimal surgical margin.Methods:By using surface-enhanced laser desorption ionization time-of-flight mass spectrometry(SELDI-TOF-MS) to analysis four cases with PTla renal clear cell carcinoma lesions of the cancer center,cancer-adjacent 0 to 0.5 cm,cancer-adjacent 0.5 to 1 cm and normal renal tissue,to research the differences in protein expression.Results:Three different protein peaks detected were molecular weight(MW) 13743.9Da,13942.5Da and 16952.7Da in Cancer Center and normal renal tissue lesions, its protein content were significantly different(P<0.001).The protein MW 13743.9Da,13942.5Da in the cancer center and adjacent 0 to 0.5cm have high expression,in contrast to adjacent 0.5 to 1cm and normal kidney tissue without expression(P<0.001).Otherwise, the protein MW 16952.7Da in normal kidney tissue and adjacent 0.5 to 1cm have high expression,by contrast,the cancer center and adjacent 0 to 0.5cm no expression (P<0.001).Conclusion:There may be tumor infiltration or residues in adjacent 0 to 0.5 cm,but not exist in adjacent 0.5 to 1cm.Surgical margin should be 1cm far from the outer edge of tumor,at least 0.5cm,to try to avoid using tumor enucleation.
