Backgrounds:Osteoporosis is a metabolic skeleton disease which character with decreased bone mass,bone mechanics and increased risk of bone fracture. Traditional Chinese medicine complex prescription could make use of multi-method to accommodate whole body. There were superiority in osteoporosis therapeutics. Chinese herbal medicine which invigorating the kidney (Gu Ling pian) was development by determination of treatment based in pathogenesis obtained through differentiation of symptoms and signs. It has great curative effect in clinical. This topic to observe the changes of Chinese herbal medicine which invigorating the kidney (Gu Ling pian) on osteoporotic ovariectomized (OVX) rat model from two,three dimensional structure and biomechanical abilities, and approach the effects of bone strength on Chinese herbal medicine.Objectives:To investigate the histomorphology effects,three dimensional structure and biomechanical abilities of Chinese herbal medicine which invigorating the kidney (Gu Ling pian) on ovariectomized rat. To explore the therapeutics mechanism of traditional Chinese medicine and offer the experiment basis for clinical practice for treating and preventing osteoporosis.Methods:1. According to the prescription of Gu Ling Pian, the liquid of Gu Ling Pian was made, which made of rhizoma drynariae( Pilous antler and some other Chinese crude drug of invigorating the kidney). The medical materials we fed to rats were as the same as which used by people. The most active component was extracted by alcohol and water and then it was made to 1 gram crude drug per milliiter by filter enrichment. This liquid were offered by traditional Chinese medicine preparation department of Southern Medical University.2. 36 three-months Sprague Dawley SPF female rats were used. Rats were randomly divided into five groups which contains control group (sham-operated group), model group, Gu Ling Pian(GLP) of high dozes group (HDG), middle dozes group (MDG) ,low dozes groups (LDG) and ethynylestradiol treated groug.3. Castrated operation was adopted to establish rat models of osteoporosis.4. One month after operation, each group was fed the same volume of physic liquor. The sham-operated group and model group were fed with normal sodium. The three GLP groups were fed with GLP (high 3.54 g.kg-1.d-1, middle 2.66 g.kg-1.d-1, low 1.77 g.kg-1.d-1, respectively). Each group's administration were conform with the equivalent dose of clinical.5. The undecalcified third lumbar vertebra sections were cut for bone histomorphometry analysis.6. The three-dimensional parameters were measured with indentation testing. 3D reconstruction was presented from serial images.7. The biomechanical properties were measured with indentation testing.8. The test of significance is worked out by the software package of SPSS 11.5. The results are expressed by (X|-)±S. Data were analyzed with Oneway-ANOVA. When the mean square was regular, LSD method was used among groups' comparison. Significance was expressed by P<0.05.Results:1. Four months after establishing the rat models of osteoporosis, compared with the shame-operating group, significant reduction of the Percentage of Tb.Ar,Tb.N,Tb.Th and significant increase of Tb.SP,BFR/BS,BFR/BV,BFR/TV,MAR,Oc.N/mm was found in the OVX rats.2. The result of histomorphometry show that, GLP and ethynylestradiol treated grougs have significant decrease on Tb.SP, BFR/BS,BFR/BV,Oc.N/mm. MDG, HDG and ethynylestradiol treated grougs have significant increase on %Tb.Ar, Tb.Th, Tb.N. Especially the MDG more significant increase than ethynylestradiol treated groug.3. The results of 3D- reconstruction show HDL has significant increase in Volume Percentage, and GLP treated groups have significant increase in Volume Voxels. Compared with the ethynylestradiol treated groug, HDG has significant increase in Volume Voxels. The three-dimensional parameters have positive correlation with %Tb.Ar,Tb.Th,Tb.N, have negative correlation with Tb.SP. There is no significant change in Volume Percentage by 3D- reconstruction.4. In Vito dynamics, the model group has significant decrease in rigidity and max-load. Treated by GLP, there are significant increase in biomechanics, but quiet alter by the ethynylestradiol treated groug.Conclusions:Chinese herbal medicine which invigorating the kidney can restrain bone absorption and promote bone formation , which may decrease the bone turnover to improve bone quality. There are conspicuous effects in microstructure of bone trabecula , especially in structure compactedness from Gu Ling Pian high group. The significance of Chinese herbal medicine on increase bone strength is conspicuous and deserved.
