Objective:To evaluate the effect of TM5-FuNC targeting to treat transplanted human liver cancer in nude mice based on internal magnetic field built inside the tumor; And to study the effect of TM5-FuNC on expression of bcl-2/bax protein in nude mice with transplanted human liver cancer.Method:1. Established human hepatoma xenograft on nude mice. 32 modes bearing cancer were divided equally into four groups on random including: group A received placebo; group B received pure 5-fluorouracil administered via tail vein injection at 25mg/kg on consecutive five days; group C received pure magnetic stent without the applying of magnetic nanoparticles; group D consists of internal magnetic field built by magnetic stent wires inserting into tumor tissue and received TM5-FuNC administered via tail vein injection at 250mg/kg on consecutive five days. The tumor was measured on the day of first treatment and 1d, 4d, 7d, 10d and 13d after 5d's treatment. Tumor tissues were observed by transmission electron microscope after observation periods.2. The expression of bcl-2, bax were examinated by immunohistochemistric method (SABC).Results:1. The volume was markedly lower in both group B (5-fluorouracil) and D (TM5-FuNC and magnetic field created inside the tumor tissues group) than those in group A (placebo), group C (pure magnetic stent without the applying of magnetic nanoparticles) (P <0.01) and the volume was markedly lower in group D than those in group B(P <0.01).2. The expression of bcl-2 protein markedly decreased more whereas the expression of bax protein increased more in both B (5-fluorouracil) and D (TM5-FuNC and magnetic field created inside the tumor tissues group) than group A (placebo), group C (pure magnetic stent without the applying of magnetic nanoparticles) (P <0.01). The expression of bcl-2 protein markedly decreased whereas the expression of bax protein was markedly increased more in group D than in group A (P <0.01).Conclusions:1. The targeted therapy of TM5-FuNC can improve the chemotherapeutic effect of 5- Fu.2. The targeted therapy of TM5-FuNC decreases the expression of bcl-2 gene, increases the expression of bax gene; therefore, it up-regulates apoptosis of the liver cancer cells and inhibits the growth of hepatocellular carcinoma.
