Objective : To investigate the changes of expression of caveolin-3 mRNA in vascular smooth muscle cells treated by PHE,PMA,PHE/BAPTA,PHE/PZ and control.Methods: Aortic vascular smooth muscle cells of Westar male were divided into five groups: group PHE, group PMA, group PHE/BAPTA, group PHE/PZ, control. Before treated cells were maintained in serum-free DMEM for 12 hours, the cells were treated and cultured in serum-free DMEM for another 12 hours .Using semi-quantitative reverse transcription-polymerase chain reaction(PT-PCR) to determine the expression of caveolin-3 mRNA with GAPDH as internal reference.Result:Compared with PHE/BAPTA, PHE/PZ and control group ,the difference of caveolin-3 mRNA in group PHE,PMA is significant(P<0.01);the difference of the expression of caveolin-3 mRNA in group PHE and PMA is significant(P<0.05);and there is no significance among group PHE/BAPTA, PHE/PZ and control(P>0.05).Conclusion: The expression of caveolin-3 mRNA of vascular smooth muscle cells took an important role in Atherosclerosis, possibly via the alterations of intracellular Ca2+ and protein kinase C.
