Background:VEGF as a protein of secreting type , which is regulated by upstream signal pathway about the transcription and expression, and the pi3k/akt signal pathway has a important role in this way.Objective To investigate the impact of phosphatidylinositol 3-kinase ( PI3 K)/Akt signal pathway in the regulation of transcription of vascular endothelial growth factor(VEGF) in lung adenocarcinoma A549 cell line .Methods The specific inhibitor of PI3K/Akt (LY294002) was used , VEGF mRNA was detected through semiquantitative reverse transcriptional polymerase chain reaction (RT- PCR) and VEGF protein was detected through ELISA kit .Results The LY294002 could reduce the VEGF protein expression of lung adenocarcinoma A549 cell line during some extent ranges . And at the different level of 10uM, 20uM, 50uM, 100uM, the level of VEGF protein corresponding of 35.4±5.1pg/ml,26.2±4.2 pg/ml, 16.5±1.9 pg/ml,9.8±1.9 pg/ml, which has significantly reduce along with the increase level of ly294002 (P<0.05).Conclusion The transcription and expression of VEGF of lung adenocarcinoma A549 is regulated by PI3K/AKt in a certain degree.
