ObjectiveTo explore the correlation between serum level of Interleukin-8 (IL-8)and Matrix metalloproteinase-9 ( MMP-9 ) in patients with non-small cell lung cancer(NSCLC) and the relationship between serum level of Interleukin-8(IL-8), Matrix metalloproteinase-9 (MMP-9) and clinicalpathological features . To evaluate the function of IL-8 or MMP-9 as tumor markers in diagnosis and metastasis of NSCLC.MethodThe level of IL-8 and MMP-9 was detected by ELISA in 141 non-small cell lung cancer patients, 40 healthy adults and 40 patients with benign pulmonary disease.ResultsThe serum level of IL-8 and MMP-9 in non-small cell lung cancer patients was significantly higher than that of healthy adults and patients with benign respiratory disease ( P < 0. 01),and the serum level of IL-8 and MMP-9 is gradually increased with the increasing of clinicopathological stage. The serum level of IL-8 and MMP-9 in non-small cell lung cancer patients with larger tumor size, lymph nodes and distant metastases was higher than that of patients with smaller size and no-metastases . But no significant difference was recognized statistically between serum level of IL-8 ,MMP-9 and histological type, differentiation grade, location of primary tumor, also no difference with gender, age, smoking history , family tumor history of non-small cell lung cancer patients . There is an obvious correlation (r=0.526) between IL-8 and MMP-9 in the serum level of non-small cell lung cancer patients . However, IL-8,MMP-9 are not good serum tumor markers in diagnosing the early stage of NSCLC because of the AUC (Area Under Curve) of IL-8 as a marker in diagnosis of NSCLC is 0.618 and MMP-9 is 0.540, but they are the valuable auxiliary parameters in diagnosing lymph node and distant metastases of NSCLC . The area under curve of IL-8 or MMP-9(AUC IL-8=0.847,AUC MMP-9= 0.805)in diagnosing lymph node and distant metastasis was bigger than that in diagnosing other items , the sensitivity of combination detectionof IL-8 and MMP-9 was 62.8 % in diagnosing lymph node and distant metastasis of non-small cell lung cancer (NSCLC) , the specificity is 92.7%, with a higher accuracy than single marker assays.ConclusionThis study has confirmed that there is a relationship between serum level of IL-8,MMP-9 and the advance of non-small cell lung cancer . There is an obvious correlation (r=0.526) between IL-8 and MMP-9 in the serum level .Serum level of IL-8,MMP-9 were valuable auxiliary parameters in diagnosing lymph node and distant metastases of NSCLC with good sensitivity and specificity by ELISA . The combination detection of serum IL-8,MMP-9 has a higher accuracy than single marker assays.
