Apoptosis is a highly organized, energydependent program by which multicellular organisms eliminate damaged,superfluous,and potentially harmful cells.After acute myocardial infarction, a series of structural changes (expansion and thinning of the infracted area) occur. Although these changes help to maintain the cardiac function at the early phase of MI, when they are not controlled, it might lead to left ventricular (LV) dysfunction.. Cellular and molecular mechanisms contributing to the LV dysfunction are not fully understood, apoptosis, both in the infarct and healthy myocardium has been shown as an important role. Preventing and reversing the myocardial apoptotic and ventricular remodeling becomes the important turning point in clinical treatment. Recently research have reported that Bcl-2,Bax and OPN correlate compactly with myocardial apoptotic after AMI. There are empirical studies and clinical applications showed that the Yindan Xinnaotong Capsule can improve myocardial ischemia, increase coronary flows, and expand the constrictive coronary vessels, but there was not enough research on myocardial apoptotic after acute myocardial infarction.This study was investigate the effects of Bcl-2,Bax,OPN on myocardial apoptotic and the effects of"Yindan Xinnaotong Capsule"on myocardial apoptotic after acute myocardial infarction in rats. Methods: AMI was established by ligating of the left anterior descending coronary artery in WS rats. 24 hours after the procedure, the surviving rats were separated randomly into four groups: AMI group,high dose of"Yindan"group,low dose of"Yindan"group,captopril group, sham-operated rats were selected randomly as non-infarction control. The cardiac muscle samples were obtained four weeks after AMI to detect the expression of Bcl-2,Bax,OPN mRNA in noninfarcted zone by reverse transcription polymerase chain reaction (RT-PCR), then the rats were sacrificed to determine infarct sizes and Apotosis index,Myocardial cell and interstitial tissue were observed by HE stain and Masson stain.Results: Compared with those of sham-operated group, the lnfarct sizes and Apotosis index, and the expression of Bax,OPN mRNA were all increased significantly(P<0.01) in AMI group after 4 weeks. Compared with AMI group those parameters were all decreased obviously in treatment group, the high dose of"Yindan"group decreased significantly, and there were no obvious differences between low dose of"Yindan"group and captopril group.Conclusions:"Yindan Xinnaotong Capsule"Could obviously Reducing the expression of Bax,OPN in myocardial tissue, by which reduces myocardial apoptotic after AMI in rats.
