The chemical composition of traditional Chinese medicine is the material base of pharmacodynamices, and is also the key to the modernization of Chinese medicine. It has been developing in recent years that Chinese medicine treat osteoporosis instead of western medicine to reduce the toxicity."Tong Gu Luo" is used to treat osteoporosis as a prescription of traditional Chinese medicine, which is composed of Epimedium, Dodder Seed ,Polygonum , multiflorum and Drynariae. In order to determine the main chemical components of each flavor of herb in "Tong Gu Luo", every herb of them was extracted with water or ethanol solution, and then through column chromatographic separation respectively. Four crude extracts (A, B, C and D) were prepared. The four extracts were extracted by ultrasonic extraction method. Ethyl acetate, 95% and a certain concentration of ethanol solution were used as extract solvent. Three chemical parts were obtained from each crude extract. At last, twelve chemical parts (A1, A2, A3, B1, B2, B3, C1, C2, C3, C4, D1, D2, D3 and D4) were obtained. Compounds in each part were analyzed using ESI-MSn. The experimental results show that the part A1 consists essentially of epmedin C, baohuoside VI, hexandraside D, and higher percentage of icariin. The part A2 consist chiefly of quercetin, icaritin, baohuoside I, and high content of icarii and sagittin.The part A3 comprise icaritin, baohuoside I, and higher percentage of sagittin. The part B1 are composed of 2,3,5,4 , -tetrahydroxyl diphenylethylene-2-O-β-D- glucopyranoside, and higher percentage of emodin and chrysophanol-8-O-β-D- glucopyranoside. The part B2 consist mainly of emodi, 2,3,5,4,-tetrahydroxyl diphenylethylene-2-O-β-D- glucopyranoside and high content of 2,3,5,4′-tetrahy-droxysilbene-2,3-O-β-D-glucopyranoside and emodin-8-O-β-D- glucopyranoside. The part B3 consist mainly of n-butyl -β-D-fructopyranoside and a higher percentage of 2,3,5,4'-tetrahydroxyl diphenylethylene-2-O-β-D- glucopyrano- side. The part C1 is composed of astragalin, luteolin, kaempferol, and higher percentage of quercetin. The part C2 consists mainly of quercetin and higher percentage of hyperin. The part C3 consists mainly of the compound which has the skeleton with kaempferol. The part D1 and D2 aren't known due to bad ESI-MS spectra. The D4 consist mainly of 5,7,3',4'-tetrahydroxyl flavanone-7-O-α-L- rhamnosyl-(1→2)-β-D-glycoside.In this paper, we have analysized the main chemical components of each flavor of herb in"Tong Gu Luo". The research had laid the foundation for studying the relationship between chemical composition and effect, identifying the effective constituents of this prescription to cure osteoporosis.
