Objective To study the effect of proliferation inhibition and induction of apoptosis of the A549 cell line of lung cancer and human umbilical vein endothelial cell ECV304 by gold nanoshell particles of human recombinant endostatin(Gold Silica Nanoshell-rhEndostatin,G-rhES) under near-infrared radiation of 820nm.Method Using self-assembly-chemical plating method to prepare gold nanoshell particles,modify the surface of gold nanoshell particles,and connect rhEndostatin, with the bond A-U to synthesis G-rhES.Detection of inhibition of cell proliferation of G-rhES of A549 cells and ECV304 cells by MTT assay;to detect cell apoptosis with Annexin-V/PI double staining method by flow cytometry(FCM);to observe changes in Cell morphology by atomic force microscope(AFM)Results(1) using self assembly-chemical plating method to prepare silicon oxide nanoshell particles coated on the surface for effective and complete payment shell, through its surface modification to connect rh-ES,polyethylene glycol(PEG) closed shell non-specific sites of the surface layer to increase biocompatibility of G-rhES; (2)The significant inhibition of proliration of ECV304 cells by rh-ES in a time-and dose-dependent manner was observed;inhibition of proliferation of A549 cells by rhES had a certain curb the trend.Compared with the cells of 549 and ECV304 without exposure in the in the near-infrared,Gold Silica nanoshell had inhibition of cell proliferation trends in the 820nm near-infrared irradiation,and it has a significant difference,G-rhES in the 820nm wavelength near-infrared irradiation on inhibit the growth of ECV304 cells and A549 cells in a dose and time dependent manner;and G-rhES in the near-infrared irradiation has a synergistic effect with cisplatin on A549 cells;(3) The apoptosis of graphics show that rh-ES can promote apoptosis of the ECV304 cell and A549,cell,and we also know the apoptosis of graphics show that Silica nanoshell not only can promote apoptosis of the ECV304 cell and A549cell, and also directly kill ceils in dose-dependent manner;(4) atonic force microscope detect the ECV304 cells treated with rh-ES did not change significantly,the overall framework of cells still exist,pseudopodium decrease over the previous,smaller holes in the surface appears.Under irradiation in the 800 nm NIR,ECV304 and A549 cells treated with gold nanoshell particles and G-rhES,the framework of cells significantly undermine the overall structure,the hole of the cell surface size uneven,Cytoplasm outflows and pseudopodia disappear.Conclusion The self assembly-chemical plating method successfully synthesize gold nanoshell particles,through their surface modification and successfully connect the drug targeting rh-ES,rh-ES to inhibit ECV304,and a time-and dose-dependent manner;rhEndostatin inhibition of A549 cells;Only under irradiation in the 820nm NIR,Gold nanoshell particles show a killing effect on ECV304 and A549 cell,the mechanism may be related to heat-induced by near-infrared absorption of apoptosis and kill cells directly;under none irradiation in the NIR,Gold nanoshell particles havn't show inhibitory effect on cell proliferation,indicating that gold nanoshell particles have good biocompatibility.Under irradiation in the 820nm NIR gold nanoshell particles have synergies with cisplatin,which may be related to hyperthermia increase cytotoxicity of chemotherapy-related drugs.Detection of the local structure show rh-ES had targeted the role of surface receptors may play its role; under irradiation in the NIR,Gold nanoshell particles and G-rhES show non-specific effect on ECV304 and A549 in vitro.
