Objective:To study the effects of Panax Notoginseng Saponins(PNS) on expression of SREBP-1c mRNA in steatotic hepatocytes L02,so as to explore the possible mechanism.Methods:1.A model of 50%calf serum induced steatotic hepatocytes was established,lipid droplets in steatotic hepatocytes were observed by inverted microscope after oil red O staining.The content of ALT,AST,ALP and LDH in supernatant and TG in hepatocytes were measured by automatic biochemical analyzer.2.Avirulent concentrations of PNS on steatotic hepatocytes were detected by MTT assay.3.Steatotic hepatocytes model was established by cultured with 50%calf serum for 48h, then divided into 5 groups:control group,model group,spontaneous recovery group,low concentration of PNS group(10μg/mL) and high concentration of PNS group(50μg/mL).After 24 hours and 48hours respectively,lipid droplets were observed by inverted microscope after oil red O staining,and TG in hepatocytes were measured by automatic biochemical analyzer.4.The expression of SREBP-1c mRNA in hepatocyte was detected by RT-PCR after 24 hours.Results:1.Compared with control group,TG content in model group of different times(24h,48h and 72h) were increased significantly(P<0.05);A great deal of orange red and red lipid droplets were observed by inverted microscopy in model group after oil red O staining,but there were a few in normal control group.Compared with control group,after 24 hours,the levels of AST and LDH were increased in model group(P<0.01),while levels of ALT and ALP were not significant difference(P>0.05);After 48 hours and 72 hours,the levels of ALT,AST,ALP and LDH in the model group were increased significantly(P<0.05,P<0.01).2.Compared with model group,after 24 hours,the content of TG in spontaneous recovery group was decreased(P<0.05).Compared with spontaneous recovery group,treated by PNS for 24 hours can declined the content of TG in hepatocytes(P<0.05,P<0.01),especially in the low concentration group;Lipid droplets in spontaneous recovery group and the PNS treatment groups were decrease when compared with model group,especially in the low concentration. After 48 hours,there is no significant different between PNS treatment groups and spontaneous recovery group(P>0.05);Lipid droplets in spontaneous recovery group and the PNS treatment groups were decrease when compared with model group,but there is not obviously different the treatment groups observed by inverted microscope. 3.Compared with control group,expression of SREBP-1cmRNA in the hepatocytes of model group was upregulated significantly(P<0.01);Compared with model group,expression of SREBP-1c mRNA in hepatocytes of spontaneous recovery group were not decreased significantly(P>0.05);Compared with spontaneous recovery group,expression of SREBP-1c mRNA was decreased in hepatocytes of PNS treatment groups,especially in the low concentration group(P<0.05).Conclusion:1.A model of hepatocyte steatosis was successfully established by cultured with 50%calf serum.Cultured with normal medium can reverse hepatocyte steatosis.2.PNS can deregulate the content of TG in steatotic hepatocytes.3.There was a high expression of SREBP-1c mRNA in the steatotic hepatocytes.PNS can decrease the expression of SREBP-1c mRNA in the steatotic hepatocytes,which maybe the therapy target for it.
