ObjectiveObserving the dynamic changes of morphology and function of vascular endothelial cells induced by Ang-(1-7) and Angll to explore the protective mechanisms of angiotensin -(1-7) on Vascular endothelium injury induced by angiotensinâ…¡.MethodsHuman umbilical vein endothelial cells(ECV-304) cultivated in DMEM were randomly divided into Control group,Angâ…¡group,Ang-(1-7) group and mixed group.After 6,12,24h, the morphologic changes of endothelial cells were observed with inverted phase contrast microscope.Flow cytometry was used to evaluate reactive oxygen species(ROS) in cells. Nitric oxide(NO) level in the cultured medium was measured by colorimetry.The content of PAI-1 and tPA in the cultured medium was measured by enzyme lined immunosorbent assay(ELISA).The concentration of MDA which was a lipid peroxidation metabolite was measured by Thibabituric acid.Resultsâ‘ morphologic observations:compared with the control group,The cells of Angâ…¡group contracted,the width of intercellular spaces increased with time went on,even necrosis and defluvium of ECs appeared;the cells of Ang-(1-7) group had no differences compared with the control group;the mixed group had less traumatic changes,the necrosis and defluvium of cells were not so obvious.â‘¡function changes:The content of ROS,MDA, PAI-1 in Angâ…¡group was higher than the other three groups;the content of ROS,MDA, PAI-1 in Ang-(1-7) group had no differences compared with the control group;the content of ROS,MDA,PAI-1 in mixed group decreased,and had statistical significances compared with Angâ…¡group.The content of NO in Angâ…¡group was lower than the other three groups;the content of NO in Ang-(1-7) group had no differences compared with the control group;the content of NO in mixed group increased significantly,and had no statistical significances compared with the control group.The content of tPA in all groups had no differences.Conclusion Ang-(1-7) had protective effects in morphology and function in endothelial cell injury induced by Angâ…¡.
