Objective:To prove the effect of compound capsule better than that of BYHWT on stress cerebral ischaemia from the anti-oxidation and neuronal apoptosis via animal experiment.With the living SD rats model of cerebral ischemia reperfusion injury,this research study the effects of BYHWT and compound capsule on pathologic change of neuronal structure in hippocampal gyrus CA1 area,neuronal apoptosis.Detectived the activity of SOD,the level of MDA in serum and brain tissue,water in brain.To observe the acute toxicity using KM mouse.Methods:1.Pharmacodynamics experiment:90 rats were divided into 6 groups at random:sham group,control group,BYHWT group,compound capsule high,middle,low dosage groups,each group had 15 rats.Rats of BYHWT group,compound capsule high,middle,low dosage groups were fed with corresponding mecidine,others were fed with equivalent physiological saline.Transient rat global cerebral ischemia was induced by modified four-vessel occlusion method.All rats were singed up bilateral vertebral artery with electric cautery.After 24hours,under the conditiong of rats were waked,rats of medicine groups were blocked bilateral common carotid artery with artery clip for 15min,then reperfused.Others were only exposed bilateral common carotid artry.The eleventh day,all rats were got blood then chose 6-7 rats from each groups at random and removed brain tissue.Half of brain was homogenated to test biochemical indicators.The other half was immobilized to study the pathologic change of neuronal structure in hippocampal gyrus CA1 area,neuronal apoptosis.2.Acute toxicity experiment:20 female and 20 male KM mice each were divided into two groups,compound capsule group and control group,each had 10 mice.The medicine groups were fed with compound capsule solution in its peak concentration and volume by gavage,the control groups were fed with equivalent physiological saline.24h and 1 week after the gavage observed the animals' behavior,drink and food,diarrhoea,death;pelage,urine and stooland their main organs' appearance.Results:1.In BYHWD group and compound capsule groups of high,middle and low dosage-treated rats,and the pathologic change of neuronal structure of neuronal cells in CA1 region were improved.2.Compared with control group,BYHWD group and compound capsule groups of high,middle and low dosage,the activity of SOD in serum increased and MDA level in decreaed apparently.And the middle dosage compound capsule group worked better than other groups.3.Compared with control group,BYHWD group and compound capsule groups of high,middle and low dosage,the activity of SOD in brain tissue increased and MDA level in brain tissue decreaed apparently.4.Compared with control group,BYHWD group and compound capsule groups of high,middle and low dosage,the content of water in brain tissue decreased apparently.And the low dosage compound capsule group worked better than other groups.5.Acute toxicity experiment showed that compound capsule has no toxic,with a good safety.Conclusion:The effect of compound capsule is better than that of BYHWD on stress cerebral ischaemia,and its mechanism may relate with increasing the activity of SOD and decreasing the content of MDA and then strengthens the brain tissue's anti-oxidant ability,lessen the damaged caused by oxyradical. Acute toxicity experiment showed that compound capsule no toxic,with a good safety.
