Objective:To evaluate the effects of the subconjunctival injection of bevacizumab on angiogenesis in alkali burn corneas of rats and compare the curative effect of the drug delivered at different times after lesion to discuss the ideal medicine time.Methods:The corneal neovascularization(CNV) model was induced by alkali burns in corneas of 36 Wistar male rats.And then they were divided in three groups(group have 12 rats and 12 corneas):control group,experimental group 1 and experimental group 2.The experimental group 1 received subconjunctivally 0.02 ml of bevacizumab(25mg/ml) at day 1,3 and 5 after lesion;the experimental group 2 received subconjunctivally 0.02 ml of bevacizumab(25 mg/ml) at day 3,5 and 7 after lesion;while the control group received subconjunctivally 0.02 ml of 0.9% saline solution at day 3,5 and 7 after lesion.All alkali burn eyes of rats received topically ofloxacin eye drops four times a day 24h after lesion.The angiogenesis and inhibition of corneal neovascularization(CNV) were detected respectively by measuring the length and the area of CNV under slit lamp microscopy,takeing count of the microvessl under optical microscopy and immunohistochemical detection of vascular endothelial growth factor(VEGF).Results:1.Observation under slit lamp microscope Compearing with control group the CNV occurred later and grew more slowly in the two experimental groups.The area of neovascularization of the corneal sureface in the two experimental groups was smaller than in control group[(66.90±12.57)%],and the difference has statistical significance(P<0.001) In addition,there was statistical differences between experimental group 1 and experimental group 2[(27.70±12.60)%vs(41.60±12.70)%,P<0.05].2.The microvessl number of the two experimental groups were few less than the control group(8.13±1.57,P<0.001).In addition,the experimental group 1(2.02±1.17) were less than the experimental group 2(3.16±1.36,P<0.05).3.Detected the immunohistochemical staining of VEGF,the expression of VEGF markedly increased in control group(126.28±7.33) compared with the two experimental groups(P<0.001).There was also statistically significant difference between experimental group 1(101.53±6.80) and experimental group 2(108.67±4.74,P<0.05). Conclusion:1.Subconjuntival injection of bevacizumab could inhibit angiogenesis and reduce the expression of vascular endothelial growth factor(VEGF) in alkali burn corneas of rats.2.Bevacizumab could also inhibit the angiogenesis that has been formed,and the sooner drug delivery,the more obvious inhibitory effect.
