Objective:To observe the expression of NF-κB,IL-1βin different times after 7 d exhausted swimming exercise , At the same time to investigate the mechanism of activition and regulation of NF-κB signaling pathway in CNS and the relationship between NF-κB signaling pathway and the inflammatory reaction of IL-1βinduced.Methods: seventy healthy SD rats were randomly divided into control group(n=10) and exhausted swimming group(n=60). The fresh cerebral tissues were divided on icewater bath while the paraform-perfused brains were obtained at 0 h, 4 h,8 h,12 h, 16 h, 24 h after seven-days exhausted exercise, the former to investigate the changes of NF-κB in amygdaloid nucleus with western-blot method, the latter to observe the expression of NF-κB and IL-1βby immunohistochemical SABC techniques.All of the experimental data were analyzed by SPSS13.0 statistical software,and expressed by (Mean±S.E.).Difference was considered statistically significant at p<0.05.Results:(1)Western-Blot results:compared with control group,the expression of NF-κB in amygdaloid nucleus increased at 4 h(p<0.05),and peaked at 8 h(p<0.05), decreased at 12-16 h(p<0.05),and returned to normal levels at 24 h(p>0.05). Immunohistochemistry results: the expression of NF-κB was extremely lower and mainly resided in endochylema in control group and 0 h group,no difference was found between them.Increased and transfered into nucleus at 4 h after exhausted exercise(P< 0.05), and reached peak levels at 8 h(P< 0.05),and decreased at 12 h(P< 0.05),16 h(P< 0.05),24 h(P> 0.05) gradually.Compared with control group,the number of IL-1βpositive cells increased in all exercise groups dramatically(P< 0.05),exclude 12 h group reached peak levels (P< 0.05),there was no significant difference in other exercise groups(P> 0.05).Conclusion: (1)These data suggest that the NF-κB signaling pathway can be activated in amygdaloid nucleus in rats by exhausted swimming exercise-induced ROS and play an important role in maintaining cellular oxidant-antioxidant balance and regulating the function of neuroendocrine system.(2)NF-κB signaling pathway and IL-1βinvolved in reversible inflammatory reaction of brain .
