1. Objective:Establish animal model for stress urinary incontinence in rats. Investigated the expression changes of Tgfβ-3,Lamin, Relaxin in paries anterior vaginase and levator ani muscle between SUI rats and normal rats .We analysis the expression of three factors between SUI and normal rats.Initially explore the mechanism of incidence of SUI animal model and three factors changing expressiong in SUI and normal animal.2. Method:Fifty female SD rats were separated into 5 groups. GroupⅠ(10 rats) was the control group. GroupⅡ(10 rats) was the group of normal childbirth. GroupⅢ(10 rats ) was the group of normal childbirth + colpectasis. GroupⅣ(10 rats ) was the group of normal childbirth + ovariectomy. GroupⅤ(10 rats) was the group of normal childbirth + colpectasis + ovariectomy. Cystometry, modified leak point pressure measurement was used to observe the effect. The expression of Tgfβ-3,Lamin and Relaxin mRNA in paries anterior vaginase and levator ani muscle was determined by RT-PCR, the level of oxidative stress was assessed by colorimetric analysis.3. Results:3.1 The values of abdominal leak point pressure (ALPP) of five groups were (74.24±5.01), (70.81±3.06) , (52.62±3.63), (53.26±3.45), and (35.20±3.61) cm H2O respectively. The values of ALPP in groupⅡwas lower than that in groupⅠ,but there was no significant difference (P>0.05). The values of ALPP in groupⅢ,Ⅳ,Ⅴwere lower than in groupⅠ,and the difference was obviously significant (P <0.05 ). The values of ALPP in groupⅢ,Ⅳ,Ⅴwere lower than in groupⅡ, and the difference was obviously significant (P <0.05 ). There was no significant difference of ALPP between groupⅢand groupⅣ(P>0.05). There was significant difference of ALPP between groupⅤand groupⅡ,Ⅲ,Ⅳ(both P <0.05).3.2 The expression of Tgfβ-3 , Lamin in paries anterior vaginase of groupⅤ(SUIgroup)was significantly lower than groupⅠ(normal group) (P <0.05). The expression of Relaxin in paries anterior vaginase ofⅤgroup (SUIgroup)was significantly higher thanⅠgroup(normal group) (P <0.05). 3. The expression of Tgfβ-3,Lamin in levator ani muscle of groupⅤ(SUIgroup) was significantly lower than groupⅠ(normal group) (P <0.05 ). The expression of Relaxin in levator ani muscle of groupⅤ(SUIgroup) was significantly higher than groupⅠ(normal group) (P <0.05 ).4. Conclusions:4.1 The animal model of stress urinary incontienece can be obtained successfully through imitating birth trauma or ovariectomy.4.2 The expression of Tgfβ-3,Lamin in paries anterior vaginase and levator ani muscle of SUIgroup was significantly lower than normal group (P <0.05 ),and also the expression of Relaxin in paries anterior and vaginase levator ani muscle of SUIgroup was significantly higher than normal group (P <0.05 ), the three factors may play an important role in the incidence of SUI.
