Expression Of Rock-Ⅱ Kinase In Ureter Smooth Muscle And Its Significance After Rabbit Partial Unilateral Ureteral Obstruction

Objective Apply rabbits partial unilateral ureteral obstruction model (PUUO),to Investigate the expression of Rock-â…¡kinase at ureteral smooth muscle tissues and its significance.Methods: Use the improved ureteral casing method to establish PUUO model.Detection ureteral resting pressure by urodynamic apparatus. Immunohistochemistry method was used to detect the Rock-â…¡kinase expression in ureteral smooth muscle.Carry out kidney and ureter morphology observation.Select meaningful experimental groups based on the above test results,to make correlation analysis of the Rock-â…¡kinase IOD(the integrated optical density)value and ureteral resting pressure.Results: 1.Rock-â…¡kinase immunohistochemistry:Rock-â…¡kinase expression in the ureteral smooth muscle cytoplasm of all groups.After 2,3 weeks of obstruction,the positive expression area and IOD of the expression of Rock-â…¡kinase in ureter smooth muscle was obviously higher than in the controls,P<0.01.The expression in 3-week group is peak.After 4 weeks of obstruction,the expression of Rock-â…¡kinase were lower than in the control groups<0.05.2. Ureteral resting pressure:after 3 weeks of obstruction, there was a higher peak value in resting pressure,then delined in 4 weeks.After 2,3weeks of obstruction,ureter resting pressure was higher than in the control group,P<0.05,the difference has statistical significance.3. Ureteral smooth muscle HE observed:After 2,3-week of obstruction,ureteral smooth muscle smooth muscle cells markedly increased,muscular layer thickening. After 4 week of obstruction,the smooth muscle cells reduce,muscular layer significantly thinning.Kidneys, ureter macroscopy: morphological changes in the kidney and ureter of all the PUUO rabbits consistented with which emerged in the literature4.Correlation analysis:In obstructive 3-week group,by the Pearson correlation test,there is a positive correlation between Rock-â…¡kinase IOD and ureter resting pressure.(r=0.677,P<0.05).In obstructive 4-week group,by the Pearson correlation test,there is a positive correlation between Rock-â…¡kinase IOD and ureter resting pressure.(r=0.653,P<0.05).Conclusions: This study showed that:(1)Rock-â…¡kinase was involved in the the development of the PUUO disease.(2)Rock-â…¡kinase also plays an important role in ureteral early compensatory mechanism.(3)After 4 weeks of obstruction, Rock-â…¡kinase expression was in low expression. Because of the role of smooth muscle morphology reduced,Rock-â…¡kinase should be intervened.(4)Rock-â…¡kinase which can affect ureteral resting pressure may be play an important role in ureteral function regulation, intervention of Rock-â…¡kinase has some significance in ureteral function.
