Objective To investigate the protective effective and possible mechanism of L—carnitine on experimental chronic hepatic injury rats induced by CCl4 on experimental acute hepatic injury rats and mice induced by CCl4 and on experimental acute hepatic injury mice induced by alcohol and APAP.Metheds To duplicate the chronic liver injury models of the rats induced by CCl4,the acute liver injury models of the rats and mice induced by CCl4,the acute liver injury models of mice induced by alcohol;and APAP respectively. To divide the rats and mice into different groups at random according to their weight: normal groups,model groups,medicine groups and LC groups(the doses and groups were diffrent according to the diffrent models).To give the CCl4 or alcohol or APAP to duplicate the models,and determin the values of ALT,AST,T-BIL,TP,ALB,A/G,calculate the liver indexes,detect pathologic change of the chronic liver injury models of the rats induced by CCl4.To detect the contents of protein and the activities of SOD,MDA,GSH-px,T-AOC,CAT.Results Duplicate the models successfully.As for the the chronic liver injury models of the rats induced by CCl4,LC of 0.15g·kg-1 doses can reduce the values of ALT,AST,liver index(LI),T-BIL,TP,ALB and A/G remarkably(P<0.01),and lessen the pathologic change of hepatic tissue injuried by CCl4 efficiently.As for the acute liver injury models of the rats and mice induced by CCl4,the acute liver injury models of mice induced by the alcohol;and by the APAP,the values of LT,AST,LI reduced remarkably,and present dose dependent.(P<0.05,P<0.01);but as for the acute liver injury models of mice induced by the alcohol,the values of LI were not statistically significant.(P>0.05)..And the activities of SOD,GSH-px,CAT,T-AOC increaced significantly(P<0.05,P<0.01),while the activity of MDA reduced(P<0.05,P<0.01).and presented dose dependent.Conclusion LC has a remarkably protective effect on chronic liver injury rats induced by CCl4,on acute liver injury rats and mice induced by CCl4,and on experimental acute hepatic injury mice induced by alcohol.and APAP.The protective mechanism relates to the antioxygen property of LC.
