Objective To study the effect of inhibition of cyclooxygenase-2 on the growth of tumors established by PC-3 cells in mice and to explore the mechanisms of the effect.Method The mouse prostate carcinoma models were established by inoculating PC-3 cells to subcutaneous tissue of mice.Tumors were palpable in all 16 animals 1 week after inoculation and mice were randomized to receive either vehicle(control) or NS398, 3mg./kg.body weight,intraperitoneally three times weekly.The inhibitory effect of NS398 on tumors was studied.HE staining was used to evaluate the morphological changes of prostate carcinoma tissue.Levels of VEGF in tumor was detected by Western blot.Result The mean tumor weights of 2 groups were(1.57±0.28),(0.58±0.16)g,The weight of tumors in the study group was significant less than in the control group(P<0.01) with 63.06%of inhibition rate and there was no significant difference of the body weight between 2 groups(P>0.05).Western blot analysis revealed that VEGF expression was significantly down regulated in the NS398 treated tumors.Conclusion NS398 can suppress PC-3 cell tumor growth in vivo.Tumor growth suppression might be achieved by down regulation of tumor VEGF with decreased angiogenesis.
