| Objective To explore the potentiality of intravecical citric acid induce Overactive bladder and the urodynamic changes of intravecical botulinum toxin type A(BTX-A) administration for Overactive bladder induced by citric acid in rats.Methods 30 rats were inserted into the polyethylene tube.In 7,10 and 14 days,Cystometry was performed and any changes in the bladder capacity and maximum voiding pressure ware evaluated,citric acid solution was used in 20 rats and normal saline was used in 10 rats.After 7 days were inserted into the polyethylene tube,20 rats were treated intravesically with 1mL of BTX-A(20U).Cystometry was performed before and in 2,6 days after instillation,citric acid solution was used in 10 rats and normal saline was used in 10 rats.Results①Cystometry using citric acid solution,the bladder capacity was significantly decr eased compared with the control rats(P<0.05) and no change was seen in the maximum voidin g pressure.No significant differents ware found in the bladder capacity or maximum voiding pressure in 20 rats or 10 rats.②Before and after in 2,6 days the instillation of BTX-A,th e bladder capacity using citric acid solution ware 0.41±0.43ml,0.58±0.53ml,1.02±0. 33ml,the result was of statistical significance(P<0.01);No change was seen in the m aximum voiding pressure.No significant differents ware found in the bladder capacity or maxi mum voiding pressure using normal saline.Conclusions Overactive bladder can be induced by instillation of citric acid in rats.Instillat ion of BTX-A inhibits bladder afferent nerve mechanisms by increasing the bladder capacity in a Overactive bladder rat model induced by citric acid.These findings may have important ap plication in treating Overactive bladder,particularly caused by bladder afferentnerve dysfunctio n. |
