In recent years, the incidence of Brucellosis increased each year, animals also have a single group between the outbreak of the phenomenon, which reflects the domestic animals of the prevalence of Brucellosis among in all regions.11880 sera and milk samples collected from cows and sheep in different provinces were detected by six methods, such as RBT, SAT, MRT, iELISA, cELISA and Milk iELISA. The results indicated that Brucellosis was prevalent in cow and sheep and the positive rate was different in various areas. As a whole, the positive rate of dairy cows was much more serious, followed by beef cattle. It was found that yak also infected Brucellosis in the latest round of inter-animal Brucellosis epidemiological survey. Within 3 rounds of epidemiological survey of Brucellosis in designated provinces, the positive rate in sheep was significantly lower than that in cow, which may relate to that cows are fed much longer than sheep and few were eliminated.In the present work, suspicious cow milk samples and the lung, liver, spleen of abortive fetal in an area of northwest China were collected and used to cultivate and isolate pathogens. The isolated strains were then stained by Gram staining and Kozlowski differential staining, and identified by a serial biochemical tests. Twelve pathogens were identified as Brucella spp. by using traditional methods and four kinds of PCR-based methods, namely BCSP31-PCR, VirB8-PCR, Multiplex PCR and AMOS-PCR. Among them, eleven strains were B.abortus biovar 3, one strains were B.suis biovar 1.Traditional classification method is an internationally recognized, on epidemiology and prevention of Brucellosis is very meaningful. However, due to large-scale use of antibiotics and disinfectants, a large area to carry out immunization, some of the local strains isolated from Category identification when it is difficult to identify the problem. There is a need in combination with other identification methods to improve the classification of Brucella identification techniques. Therefore, this study explored the genus Brucella classification of molecular biology, we use the MLVA to identified some reference strains,vaccine strains and isolates for Brucella spp.
