Objectives: The study investigated the associations between MDR1 C3435T polymorphism and the clinical and pathological characteristic and prognosis of non-hodgkin's lymphoma (NHL); Expression of P-glycoprotein and the clinical and pathological characteristic and prognosis of non-hodgkin's lymphoma (NHL).And the association between MDR1 C3435T polymorphism and the Expression of P-glycoprotein.Methods: This study collected a total of 67 NHL patients'peripheral vein blood and a total of 140 NHL patients'pathological section and the clinical data of these patients in GuangXi Province. The MDR1 C3435T polymorphism were genotyped by polymerase chain reaction -restriction fragment length polymorphism (PCR-RFLP) analysis. The expressions of P-glycoprotein were tested by immunohistochemistry technique. The Association between polymorphisms and P-gp and NHL were estimated by chi-square test of SPSS 13.0 statistics analysis sofeware system.The chi-square test was used to evaluate the association between MDR1C3435T poly- morphism &Expression of P-glycoprotein and recent therapeutic effect, blood toxicity, the number and size of organs invaded outside of lymph gland and serum LDH.Results: 1. the associations between MDR1 C3435T polymorphism and the clinical and pathological characteristic and prognosis of non-hodgkin's lymphoma (NHL).(1) The CR rate and remission rate have no significant differences between the(CC/ CT ) genotype and the TT genotype(P >0.05).(2) the association between MDR1 C3435T polymorphism and blood toxicity。①There's significant difference in WBC IV decrease between the(CC/ CT ) genotype and the TT genotype(P=0.042).②The hemoglobin and platelet decrease have no significant differences between the(CC/ CT ) genotype and the TT genotype(P >0.05).(3)The biggest focus of infection of NHL , the numbers of organs invaded outside of lymph gland and serum LDH have no significant differences between the(CC/ CT ) genotype and the TT genotype(P >0.05).2. The associations between the expression of P-glycoprotein and the clinical and pathological characteristic and prognosis of non-hodgkin's lymphoma (NHL).(1) The CR rate and remission rate have no significant differences between the P-gp (-) ones and theP-gp (+) ones(P >0.05).(2)The white cell, hemoglobin and platelet decrease have no significant differences between the P-gp (-) ones and the P-gp (+) ones(P >0.05).(3)The biggest focus of infection of NHL , the numbers of organs invaded outside of lymph gland ,age and serum LDH have no significant differences between the P-gp (-) ones and the P-gp (+) ones(P >0.05).3. The expression of P-glycoprotein has no significant differences between the (CC/ CT ) genotype and the TT genotype(P=0.420).Conclusions:1. The MDR1 C3435T polymorphism cann't predict the recent therapeutic effect of NHL. But the result should be verifycated in a larger sample.2.The TT genotype is easier to suffer WBC IV decrease. But it has no associations between MDR1 C3435T polymorphism and hemoglobin and platelet decrease.3.It has no associations between MDR1 C3435T polymorphism and The biggest focus of infection of NHL , the numbers of organs invaded outside of lymph gland and serum LDH.4.TheP-gp (-) ones have higher CR rate than the P-gp (+) ones.TheP-gp (-) ones may have better recent therapeutic effect, but the result should be verifycated in a larger sample.5. It has no associations between The expression of P-glycoprotein and the white cell, hemoglobin and platelet decrease.6. It has no associations between The expression of P-glycoprotein and the biggest focus of infection of NHL , the numbers of organs invaded outside of lymph gland ,age and serum LDH.7.The study don't find any causal relation between MDR1 C3435T Polymorphism and the expression of P-glycoprotein. MDR1 C3435T Polymorphism cannot predict the P-glycoprotein expression of tumor tissue.
