Purpose: Caloric restriction(CR), also known as dietary restriction(DR) is refered to limiting total calories in daily intake,but provide organisms adequate nutrients such as essential amino acids, vitamins and so on, to ensure the organisms not to occur malnutrition.With masses of biological effects,CR has become a hot spot of medical and biological study.Some animal experiment and clinical research also showed that the lipid peroxidation and enhance antioxidant enzyme activity can be ruduced by CR or exogenous antioxidant nutrients supplement.The free radical theory of medicine pointed out if human body cell suffered a long term attack of harmful free radicals,the normal metabolism of cells will be affected.Cells can not be repaired,then will be aging and suffer disease even to death. According to organism's physiological needs,selecting nutriment containing antioxidant substances will be able to prevent disease and aging.The purpose of this study is to investigate the effect of CR and curcumin to serum and hepatic SOD activity, MDA, TAOC level in adult rat, disscuss its possible mechanism, so as to provide experimental basis for human healthy diet.Methods : The studies were selected 2.5-3-month-old Sprague Dawley male rats .the total of 46 rats were randomly divided into 4 groups: groupⅠ(control group,12 rats), groupⅡ(high calorie group ,12rats),groupⅢ(CR group,12rats),groupⅣ( high calorie + turmeric group ,10rats). There was no significant difference between the beginning rat weight of each group. After fed for 90 days, the rats were killed through injecting intraperitoneally with 10% chloral hydrate and bleeding the femoral arteries, then serum and liver tissue samples were obtained for assaying activity of Serum and tissue homogenate SOD and content of serum MDA,TAOC. Also hepatic expressions of protein activity were analyzed by Immunohistochemical method. The data were analyzed using SPSS for Windows11.5Quantitative datawere expressed as ( x±s). P values of <0.05 were considered significant.Results:⑴weight change: Before feeding,body weight of rats in each group to meet homogeneity of variance,body weight of rats has no statistical significance among the four groups (p>0.05). After a month,Compared with control group. GroupⅡshowed significantly increased body weight,GroupⅢand groupⅣshowed slowly increased body weight,The differences have statistical significance (p<0.05). After two month , Compared with control group, groupⅡshowed increased body weight,the differences have no statistical significance(p>0.05),GroupⅢand groupⅣshowed slowly increased body weight , The differences have statistical significance (p<0.05). After 3 months,Compared with control group, groupⅡshowed increased body weight,the differences have no statistical significance(p>0.05),GroupⅢand groupⅣshowed slowly increased body weight,The differences have statistical significance (p<0.05).⑵Compared with control group, groupⅢshowed slightly increased in serum SOD activity which was no significant difference(p>0.05)and significantly increased activity of SOD in rat liver tissues (p <0.05). GroupⅡshowed significantly decreased activity of SOD in serum(p<0.05);and slightly decreased activity of SOD in tissues(P>0.05). groupⅣshowed slightly decreased activity of SOD in serum and tissues,but there was no significant difference(P>0.05).Compared with GroupⅡ, groupⅢshowed significantly increased activity SOD in serum and tissues(p<0.05, p<0.05); groupⅣshowed slightly decreased activity of SOD in serum and tissues,but there was no significant difference(P>0.05).the differences between groupⅢand groupⅣhas no statistical significance in serum SOD activity (p>0.05). but have statistical significance in tissues SOD activity(p<0.05).⑶Compared with control group, groupⅢand groupⅣshowed significantly decreased in serum content of MDA(p<0.05) and slightly decreased in liver tissues content of MDA (p>0.05). GroupⅡshowed slightly decreased and increased in liver tissues content of MDA (P>0.05). Compared with GroupⅡ, groupⅢshowed slightly decreased content of MDA in serum and tissues(p>0.05); groupⅣshowed slightly decreased content of MDA in serum(p>0.05), but significantly decreased in tissues(p<0.05).The differences between groupⅢand groupⅣhas no statistical significance in serum and liver tissues content of MDA (p>0.05).⑷Compared with control group, groupⅢshowed slightly decreased content of TAOC in serum but slightly increased in liver tissues(p>0.05). GroupⅡshowed significantly decreased content of TAOC in serum and liver tissues (p<0.05). groupⅣshowed significantly increased in serum(p<0.05),but there was no significant difference in liver tissues content of TAOC (P>0.05) .Compared with GroupⅡ, groupⅢshowed slightly increased content of TAOC in serum (p>0.05) and significantly increased in tissues(p<0.05); groupⅣshowed increased content of TAOC in serum and tissues(p<0.05).Compared with groupⅢ, groupⅣshowed significantly increased content of TAOC in serum(p<0.05).but there were no significant difference in tissues(P>0.05).⑸Immunohistochemical results showed that groupⅡ,control group and groupⅣhad slightly hepatic SOD expressions, but low-calorie feed group SOD expressions had increased significantly.Conclusion: Our results indicates that caloric restriction can significantly enhance activity of antioxidase SOD,thus inhibit lipid peroxidation. High-calorie diet can reduce activity of antioxidase SOD and total antioxidant power,turmeric can alleviate high-calorie-induced activity SOD decreasing ,and then reduce the lipid peroxidation product.whether or not to inhibit free radical generation ,enhance antioxidant system and reduce damage caused by oxidative strees through food adding anti-oxidant or CR,it requires further study.
