Object:Renal interstitial fibrosis is a variety of end-stage renal disease common pathology.UUO was using Preparation obstructive nephropathy model,eventually leading to renal interstitial fibrosis.Renal interstitial fibrosis formation is mediated by a variety of factors caused by,including hemodynamic factors and non-hemodynamic factors,in hemodynamic factors the renin-angiotensin system(RAS) is most closely related.RAS,TXA2,6-keto- PGF1a,plasma endothelin-1(ET-1) and serum nitric oxide(NO) also play the very important role in this procedure.Unbalance of them affect structure and function of kidney.With the cellular and molecular biology methods of development,we consider a variety of vasoactive substances,metabolic disorders,as well as the resulting hemodynamic abnormality is to promote the condition of chronic progressive development important aspect,we are concerned the changes of RAS,thromboxane B2(TXB2),6-keto - prostaglandin(6-keto- PGF1a),endothelin-1(ET-1),nitric oxide(NO) in UUO rats.In this study,we observe the effect of shenluotong on RAS,thromboxane B2,6-Keto-PGF1a,ET-1 and NO in rats with UUO.Motheds:Forty male SD rats were used in this experiment, the animals were randomly divided into Sham group,UUO group,UUOV group and UUOS group(n=10).The left ureter was ligated for this study.At the 14th day the 24-hour urine, blood were collected for testing.Kidneys were harvest for histopathological observation,renin,angiotensin-converting enzyme(ACE),AngⅡ,TXB2,6-keto- PGF1a,ET-1 and NO were measured by radioimmunoassay method.Expression of ACE was measured with immunohistochemical method.Results:l Eeffect of shenluotong on 24 hour urinary protein in UUO ratsThe 24hs proteinuria in UUO group was significantly higher than Sham group(P<0.05);compared with UUO group the treated group decreased markedly,there is significant difference between them(P<0.05).But there in no differences in treated groups.(P>0.05)2 Eeffect of shenluotong on Scr and BUN in UUO ratsScr and BUN in UUO group was significantly higher than Sham group(P<0.05);compared with the UUO group,the treated group were decreased markedly(P<0.05).3 Eeffect of shenluotong on renin angiotensin system in UUOrats3.1 The activity of renin in renal tissueCompared with the Sham group,renin activity in UUO group,UUOV group and UUOS group renal tissue were inhibited significantly(P<0.05).There is no significant difference among of them(P>0.05). 3.2 Expression of ACE in renal tissueExpression of ACE with Immunohistochemistry showed that weak expression in Sham group and re-regulated with UUO. Compared with it,UUOV and UUOS group were down-regulated significantly(P<0.05).There is no significant difference between UUOV and UUOS group.(P>0.05)3.3 The content of AngⅡin renal tissueCompared with UUO group,AngⅡin Sham group,UUOV group and UUOS group was significantly lower(P<0.05).There is no significant difference between treated groups(P>0.05).4 Eeffect of shenluotong on the content of TXB2 in UUO ratsCompared with Model group,UUOV group,UUOS group, Sham group TXB2 content in renal tissue decreased significantly (P<0.05);Compared with the Sham group,content of TXB2 in UUOV,UUOS increased significantly(P<0.05);There is no significant difference between UUOV and UUOS group.(P>0.05).5 Eeffect of shenluotong on the content6-keto- PGF1a in UUOratsCompared with UUO group,6-keto- PGF1a content in renal tissue UUOV group,UUOS group,Sham group decreased significantly(P<0.05);Compared with the Sham group, 6-Keto-PGF1a content in renal tissue in UUOV group,UUOS group increased significantly(P<0.05);There is no significant difference between UUOV and UUOS group.(P>0.05).6 Eeffect of shenluotong on the content of ET-1 in UUO rats Compared with UUO group,ET-1 content in renal tissue Sham group,UUOV group and UUOS group kidney tissue decreased significantly(P>0.05);There is no significant difference among of them(P>0.05).7 Eeffect of shenluotong on the content of NO in UUO ratsCompared with the Sham group,UUOV group and UUOS group was increased(P>0.05);Compared with UUO group, There is no significant difference between UUOV and UUOS group.(P>0.05).8 Eeffect of shenluotong on the expression of ACE in UUO rats by Immunohistochemical methodThe expression of ACE in Sham group was weak positive (0.85±0.11),The expression of ACE in the UUO group Markedly enhanced(2.65±0.18);Compared with the sham group, the expression of ACE in UUOV(1.70±0.17) group and UUOS (1.65±0.18)group has significant difference(P<0.05); Compared with UUO group,the expression of ACE in UUOV (1.70±0.17) group and UUOS(1.65±0.18)group reduced significantly,(P<0.05).There is no significant difference between UUOV and UUOS group.(P>0.05).Conclusion:1 Shen Luo Tong can alleviate the lesion of renal tissue and reduce the desposition of ECM in obstructive kidney,prolong the processes of fibrotic.2 Shen Luo Tong can regulate abnormal of RAS system in kidney,and inhibit the content of ACE and AngⅡ,and reduce damage to the kidneys.3 Shen Luo Tong can inhibit secretion of thromboxane B2 and endotheLin-1,improve renal blood circulation,inhibit cell proliferation.
