ObjectiveTo study whether cultured human trabecular meshwork cells express Angâ…¡in vitro, and to explore the way RAS participate in the regulation of intracular tension and to develop a new way for the self-ballance and regulation of intraocular tension.MethodsThe human trabecular meshwork cells were cultured in vitro and passaged 2 times,then the cells were transfered or cultured onto steriled cover glass. Immunohistochemical technique-Supervision method was used to localize the Angâ…¡protein in human trabecular meshwork cells. Western blot was applied to determine the existence of Angâ…¡protein in human trabecular meshwork cells.ResultsHuman trabecular meshwork cells showed yellow-brown staining in the membrane and cytoplasm, indicating a positive expression of Angâ…¡. There was no positive stain in negative control. Angâ…¡protein was found in human trabecular meshwork cells.ConclusionThe result implies that human trabecular meshwork cells have the ability of synthesizing Angâ…¡. It suggests that Angâ…¡may play an important role in the pathology of human trabecular meshwork cells, the regulation of intraocular tension and glaucoma.
