Objective: To investigate the maturation, fertilization and developmental competence of an immature oocytes derived from controlled ovarian hyperstimulation(COH) cycles by in vitro maturation (IVM)culture, and to approach the clinical use of IVM。Methods:The maturation rate of oocytes, normal fertilization rate, cleavage rate,formation rate of top quality embryo and Blastocyst rate between oocytes matured in vivo and in vitro were compared after fertilization by conventional IVF or ICSI. Results:72.41% GV oocytes and 76.08% MI oocytes reached to MII within 36h IVM , There was no difference in maturation rate(P>0.05). The normal fertilization rate between oocytes maturated in vivo and in vitro had no difference( P>0.05),but cleavage rate and formation rate of top quality embryo derived from oocytes matured in vitro were significantly lower when compared with oocytes matured in vivo( P<0.05). There were 13 Blastocys obtained.Conclusions:The immature human oocytes derived from COH cycles could mature in vitro. Although formation rate of top quality embryo was lower than oocytes matured in vivo. The number of transferable embryos in same cycle of COH were increased by IVM. The IVM was a viable method for immature oocytes gained from the cycle of COH. Objective:This study was to investigate the effect of 200μmol/L Roscovitine on in vitro maturation , mleiotic spindle and chromosome of human oocytes. Methods:Immature oocytes were cultured with a two-step culture system.Firstly immature oocytes were cultured in maturation medium with 200μmol/L Roscovitine 6 h then in maturation medium without 200μmol/L Roscovitine24-36h. Laser scanning confocal microscope (LSCM) was used to examine the mleiotic spindle and chromosome. The oocytes in control group were in TCM-199 maturation medium.Results :In experimental group was observed 61 oocytes ,after in vitro maturation 50 became maturation.There were 39o f it have spindle and chromosome,include 15 abmormal spindle and 11 abmormal chromosome.In control group was observed 60 oocytes ,after in vitro maturation 43 became maturation.There were 32 of it have spindle and chromosome,include 14 abmormal spindle and 11 abmormal chromosome. The maturation rate between experimental group and in control group had no difference( P>0.05).Conclusions : Roscovitine can reversibly inhibit nuclear maturation of human oocytes,.which is nod have bad effcct of occytes, spindle and chromosome, which may increase development competence in murine oocyte.
