Traditional Chinese medicine Polygonum multiflorum has the function of hair blacking . In this paper, the method of ethanol preliminary extraction of Polygonum multiflorum was used. Using B16F1 murine melanoma cells as materials and applying different concentrations of the ethanol extract of Polygonum multiflorum to stimulate the cells, MTT was used to study the proliferation of the cells, the tyrosinase activity was measured by enzymological method of L-dopa reaction in vitro. The synthesis of melanin was measured by NaOH-assay,and the method of RT-PCR was used to detect the expression of tyrosinase gene, tyrosinase-related protein -1 (TRP-l ) gene, tyrosinase-related protein -2 (TRP-2) gene and MITF. The results showed that the ethanol extract of Polygonum multiflorum could stimulate the proliferation of B16F1 murine melanoma cells, activate the activity of tyrosinase in the cells, increase the synthesis of melanin and improve the expression of the above four genes. Moreover, 2 mg/ml of the ethanol extraction seemed to be the best concentration for the proliferation of B16F1 murine melanoma cells, the activity of tyrosinase and the expression of the four genes. Promoting the expression of related genes in the process of the synthesis of melanin in cells, was probably one of the mechanisms that traditional Chinese medicine Polygonum multiflorum can black hair.With the proposition and development of the theory of the modernization of traditional Chinese medicine, using modern pharmacology to explain the functions of traditional Chinese medicine has become the necessary precondition for the modernization of traditional Chinese medicine. This paper verified the reasonableness of the function of Polygonum multiflorum to promote the synthesis of melanin with the methods and means of modern pharmacology, explained its mechanisms, strongly supported the effectiveness of Polygonum multiflorum and provided valuable references for the modern evaluation of the efficacy of traditional Chinese medicine. The experiment has also provided with the basis of preparations for the production of Chinese medicine compound, the animal experiments and human experiments which aim to verify the hair-blacking efficacy of Chinese medicine in the future.
