Objective: To investigate the expression and significance of high mobility group protein A1 (HMGA1)and high mobility group protein A2 (HMGA2)in endometrial carcinoma cell lines AN3CA, ECC-1, Ishikawa and tissue specimens of carcinomatous endometrium and normal endometrium.Methods: Reverse tanscriptase-polymerase chain reaction (RT-PCR) method was applied to detect the expression of HMGA1-mRNA and HMGA2-mRNA and Western blot analysis respectively was applied to detect the expression of HMGA1 and HMGA2 protein in endometrial carcinoma cell lines AN3CA, ECC-1, Ishikawa and tissue specimens from cacinomatous endometrium (n=21) and normal endometrium (n=18).Results: The expression of HMGA1-mRNA and HMGA1 protein was detected in all of the endometrial carcinoma cell lines AN3CA,ECC-1,Ishikawa and all of the tissue specimens of normal endometrium, cacinomatous endometrium.Higher expression of HMGA1 at both mRNA and protein level was detected in cacinomatous endometrium tissue, compared with normal endometrium tissue(P<0.05).The expression level of HMGA1-mRNA and HMGA1 protein was correlated with clinical stages of endometrial carcinoma (P<0.05) and there was no significant differences in differentiated feature of endometrial carcinoma (P>0.05).HMGA2-mRNA was detected in all of the endometrial carcinoma cell lines AN3CA, ECC-1, Ishikawa.The positive expression rate of HMGA2-mRNA in stage I and stageâ…¡tumors were 8.3% (1/12)and 88.9%(8/9),respectively.The positive expression rate of HMGA2-mRNA was correlated with clinical stages of endometrial carcinoma(P=0.000,P<0.05).The positive expression rate of HMGA2-mRNA in the highly, moderately and lowly differentiation tumors were 66.7%(4/6)and 33.3%(5/15),respectively.The positive expression rate of HMGA2-mRNA showed an insignificant difference in differentiated feature of cancer cells(P=0.331,P>0.05).HMGA2-mRNA was not detected in all of the tissue specimens from normal endometrium.Western blot analysis showed similar results with RT-PCR method.Conclusion: The over-expression of HMGA1 gene and the expression of HMGA2 gene may be involved in the development and tumorigenesis of endometrial carcinoma and maybe play an important role in cell proliferation and invasion of endometrial carcinoma.
