Background:Recently,the incidence of the patient damaged by electrical current has been increasing.,which has accounted for 8%of the burn patient,and its amputation rate reached as high as 64.5%.In the electrical injury cases, peripheral nerve is the most difficult recovery tissue.So the peripheral nerve recovery has become the modern electrical injury research trend recently. Voltage-gated ion channels are the structure foundations of neurons and other excitability cells to generate electrical signal,even impalpable changes on these channels' structure will affect the neural function.When the electrical injury happens,the structure will be changed,which of corresponding dorsal root ganglion(DRG)neurons membrane macromolecular proteins.Our experiment is study the changes of ion channels after the peripheral nerve was electrical injury. According to study the effect of current to membrane macromolecular protein, especially the degeneration and regeneration process of channel after electrical injury,we could see about the contribution of neuronfunctional changes.This can reveal the injury degree of nerve at protein and molecular level.From the research,we want to probe the situation of DRG ion channels after electrical injury,and master the relationship between the harms of channels after electrical injury and the disfunctions of nerves.Partâ… :Immunohistochemistrical study of dorsal root ganglion ion channel submits SCN9A,Kv1.5 polyclonal antibody in rabbit peripheral nerve after electrical injuryObjective:To study dorsal root ganglion ion channel submits SCN9A,Kv1.5 expression changes when peripheral nerve after electrical injury.Methods:The experimental animals were randomized into 4 groups according to the electrical voltage:the control group,the 50v group,the 110v group,and the 220v group.And samples were obtained instantly,4weeks.The model of the rabbit sciatic nerve with electrical injury was set up.Immunohistochemistry in semi quantitative analysis was detected the DRG ion channels expression.Results:After the electrical injury,the DRG ion channel expression is in a decreasing order,furthermore,it is inverse correlation with voltage and time.The 220v group has not obvious recovery after 4 weeks.Conclusions:The degeneration and regeneration progress of ion channel after electrical injury,depend on voltage and time.Partâ…¡:Expression changes of potassium channels and sodium channels of peripheral nerve in rabbit after electrical injuryObjective:To study the expression level of voltage-gated potassium channels and voltage-gated sodium channels after electrical injury of different voltages.Methods:The sciatic nerves of rabbits were respectively injured by 50V,100V, 150V and 200V alternating current.One week later,the mRNA expression levels of Kv1.2,Kv1.4,Kv1.5,Kv11.1,total Kv,Nav1.1,Nav1.7 and total Nav were tested as they were identified as PCR products from mRNA prepared from DRG neurons.Results:In general,the tendency of Kv1.2,Kv1.4,Kv1.5,Nav1.1,Nav1.7,total Kv and total Nav mRNA expression was decreasing;the tendency of Kv11.1 mRNA expression was increasing after electrical injury.Conclusions:The mRNA expression levels of voltage-gated potassium channels and voltage-gated sodium channels are changed after electrical injury.
