Objective To investigate the dynamic change of p53 and bcl-2 in the common carotid artery of rats after balloon injury and their effect on intimal proliferation. To evaluate the effect of all-transretinoic acid (ATRA) on intimal proliferation and the expression of p53 and bcl-2 after balloon injury. Then discuss the therapy of ATRA on restenosis after percutaneous coronary intervention(PCI).Methods The 64 male SD rats were randomly divided into three groups:sham operation group (n=8),ballon injury group (n=32,including 2, 7, 14, 28-days subgroup), ATRA administration group(n=24,including control subgroup ,normal-dose subgroup,high-dose subgroup).The balloon injury model of rat common carotid aortic artery was established.HE staining was used to observe the ratio of intima-media(I/M)thickness of the injured arteries with optical microscopy.The expression level of p53 and bcl-2 mRNA were examined with the method of RT-PCR.The expression level of bcl-2 protein was examined with the method of immunohistochemistry.Results The intimal proliferation could be found in injured arteries and the intima-media ratio was found reaching the peak(2.301±0.049 vs control 0.019±0.001,P<0.05)at 28th day.The level of p53 mRNA was found increasing after balloon injury,but bcl-2 was decreasing.The expression of p53 was observed to reach the peak at the 14th day(1.033±0.062 vs control 0.356±0.039,P<0.05),and bcl-2 was observed to reach the lowest level at the 14th day(0.826±0.035 vs control 1.5±0.029,P<0.05).The expression of bcl-2 protein was found decreasing after balloon injury,the protein was observed to reach the lowest at the 14th day(4.692±0.651 vs control 16.338 ±0.792,P<0.05).The expression of bcl-2 protein was correlated negatively with it (r=-0.769,P<0.01).ATRA inhibited,in a dose-dependent manner, the intimal proliferation and the expression of bcl-2 ,but promoted the expression of p53.The effect of high-dose ATRA was more remarkable than the normaldose(P<0.05).Conclusion The dynamic change of p53 and bcl-2 mRNA and its protein after balloon injury plays an important role in intimal proliferation.The expression of p53 and bcl-2 is correlated remarkably with intimal proliferation after balloon injury.The mechanism that ATRA suppressed intimal proliferation may involve the effect on the expression of p53 and bcl-2.
