Objective: To investigate effect of high dose methylprednisolone (MP) on heat shock protein (HSP) 27KD mRNA expression after spinal cord injury.Methods: Thirty SD rats were divided equally into operation but none injury group, Allen's weight drop injury group and injury with high dose MP group. Twenty four hours after the operation, 5 ml/kg of saline was delivered to SCI group in 30 minutes after injury. the spinal lesion and neighbor areas were moved for examination of HSP27 mRNA expression by retrograde Polymerase chain reaction technique (RT-PCR).Results: The HSP27 mRNA expression were 0.0643±0.0152, 1.0013±0.3861(P<0.01), 1.2858±0.1384(p<0.05) in three group. It increased 15 and 20 fold after spinal cord injury and treatment with MP.Conclusions: MP can greatly increase the expression of HSP27 and protect the neighbor areas after spinal cord injury.
