[Purpose]To approach stress phenomenon of endoplasmic reticulum in Alzheimer's disease and influence by statins.[Methods]Adopt lateral ventricles stereotaxis to inject with Aβ25-35 to build up rats AD model. Morris Water maze assay test rats'incubation period to observe the influence of rats different doses lipitor in incubation period. Adopt Western-Blot to test Caspase-12 albumen expression of each brain tissue simultaneously.[Result]This study indicates that latent phase among control group and therapy group decreases obviously. Compared with control group, model group rats spatial discrimination learning mnemonic dysfunction conspicuously(P<0.01); Compared with model group, huperzine A group spatial discrimination learning mnemonic improved, just the forth and fifth days showed obviously (P<0.05).But team Atorvastatin 5mg, Atorvastatin10mg, Atorvastatin 20mg groups could improve experimental AD rats spatial discrimination learning mnemonic dysfunction conspicuously obviously. Discrepancy has significance, (P<0.05或P<0.01),at the same time, the effect of Atorvastatin 10mg and Atorvastatin 20mg are higher than Atorvastatin 5mg(P<0.05) and have an advantage effect than Huperzine A group, but the two teams don't have significant deviation. . Detection of caspase-12 genetic expression adopting to Western blot and RT-PCR,control group rats brain tissue expression of caspase-12 albumen reach minimizes at the fifth day. Compared with model group, deviation has statistical significance (P<0.05); different doses atorvastatin(5mg,10mg,20mg) and Huperzine A group expression of caspase-12 albumen decrease in different level. atorvastatin 10mg reduction is extreme obviously. Compared with model group, deviation has statistical significance.[Conclusion]There is endoplasmic reticulum stress phenomenon in rats AD model。Caspase-12 protein induces stress of endoplasmic reticulum and enhances the contents of Aβ,which causes neural cells apoptosis.Different dose atorvastatin down regulate the expression of caspase-12 protein in exprimental rats AD model and obviously improves learning and memory in rats.This results indicate that atorvastatin may down regulate expression of caspase-12 protein,and thus weak stress of endoplasmic reticulum as well as neural cells apoptosis, finally improves learning and memory in rats.This may be one of mechanism of atorvastatin treating exprimental rats AD model. Offers new theory foundation and ideas for treaing AD patients.
