Purpose:Observation Liuweidihuangwan's preventive and therapeutic effect on diabetic nephropathy(DN) rats and the effects of podocyte injury.Method:1.60 SPF level male Wistar rats(weight 180-220g) will be adaptive feeding after one week,fasting but given water for 24 hours,a random check as the nine normal control group(NG),the remaining 51 by a one-time abdominal injection of 65mg/kg Streptozotocin(streptozocin,STZ).STZ before clinical use of sodium citrate buffer(0.1 mol/L,pH4.5) to 1%concentration of the preparation,spent within 10 minutes;NG injection of sodium citrate buffer contour.Detected after a week fasting blood glucose>16.7mmoL/L were regarded as into a mold,or else be removed. Again will become the mode rats were randomly divided into model group(MG) 12 only Liuweidihuangwan high dose group(HG) 15 only Liuweidihuangwan low dose group(LG) 12 only valsartan treatment group(AG) 12 only Into a mold immediately after the beginning of intervention treatment,treatment of 6 weeks. 2.HG and LG,respectively,to give Liuweidihuangwan 8,2 g/ kg·d,such as the preparation for the volume of suspension gavage; AG given valsartan 40mg/kg·d;delivery volume are 2ml/200g.NG and MG,such as the volume of normal saline given orally.3.Record each group of rats under normal circumstances.After intraperitoneal injection STZ 3 days,collection the first,fourth, sixth weekends 24h urine measured 24h urinary protein(24hupq),the sixth weekend urine testβ2-microglobulin(β2-MG),urinary albumin(ALB),urine IgG.Detect blood glucose after intraperitoneal injection STZ 3 days of the first,fourth,sixth weekend.Last 24h after treatment administration detect serum creatinine(Scr),blood urea nitrogen(Bun),albumin(ALB), triglyceride(TG),total cholesterol(CHOL);check renal tissue by immunohistochemical detection monocyte chemoattractant protein-1 (MOP-1),vascular endothelial cell growth factor(VEGF),RT-PCR detection of Podocyte' s SD nephrin,observation Pathological change by optical microscopy;detect the ultrastructural changes in electron microscopy.Through observation,comparison of the test results in each group to explore the situation Liuweidihuangwan of DN rats and the impact of podocytes' injury.Result:1.The general situation:the treatment group of rats' situation is better than MG at varying degrees.2.Clinical indicators:Liuweidihuangwan able to controlling DN rats' blood sugar,lowering its' TG and CHOL,improving lipid metabolism disorder.3.Proteinuria:Liuweidihuangwan and valsartan all can reduce DN rats' proteinuria. 4.MCP-1,VEGF:Liuweidihuangwan can significantly reduce the DN rats' MCP-1,VEGF expression,control DN rats' kidney inflammatory response.5.Podocyte' s SD nephrin:Liuweidihuangwan and valsartan all can improve nephrin mRNA expression.6.Pathology:pathology of the treatment group showed no significant rat glomerular basement membrane thickening,no significant mesangial hyperplasia,only part of foot process fusion, the degree of pathological changes in different degrees to alleviate than MG.Conclusion:The results of this study show that:Liuweidihuangwan can control DN rats' blood glucose,blood lipids,reduce proteinuria, inhibited MCP-1,VEGF expression,enhance the expression of nephrin mRNA;hinted Liuweidihuangwan can by controlling blood sugar,improving lipid metabolism,reducing DN rats' glomerular inflammatory response such variety of ways to improve the treatment of DN rats,and to some extent better than valsartan.
