| ObjectiveTo investigate the effect of pre-stimulation with the acupuncture and moxibustion on learning and memory ability and SOD(Superoxide dismutase super oxide dismutase),and NOS (Nitril Oxide Synthase Nitric oxide synthase) in hippocampal tissue of Alzheimer disease(Alzheimer disease,AD) rat model, combinated with the concept of preventive treatment of diseases, on the basis of acupuncture treatment of senile dementia currently.MethodsFirst,140 rats are taking part in Morris water maze test, excluding those who failed,and leaving 120 qualified rats weighing respectively were randomly divided into 6 groups: normal group,sham-operated group,model group,preacupuncture group,pre-moxibustion group,pre-acupuncture and moxibustion group,each has 20 rats.The 6 group rats,after three courses of treatment of 20 days,are modeled with injecting well incubated Aβ25-35 or pisiological saline into rats' hippocampus tissue.To objective the effect of pre-stimulation with the acupuncture and moxibustion on learning and memory ability with application of Morris water maze test.Finally,heart perfusied with polyformaldehyde and brain removed rapidly,in 2mm Department plane before and after the optic chiasm the hippocampus was isolated,and the visible light spectrophotometer was used to determine the changes of activity of rats' SOD,NOS inhippocampal tissue,the results using statistical treatment.Results1 Influence of pre-stimulation on Alzheimer rat models' average escape latency in channel-type water maze.Analysis froma different time period,former model,the average escape latency of each group was no significant difference(P>0.05).after model,the average escape latency significantly increased than that of former model,there is a very significant difference(P<0.01).And after model,the natural group, sham-operated group,pre-needle group,pre-moxibustion group, pre-acupuncture and moxibustion group,the average escape latency of each group comparing with former model each group itself there was no significant difference(P>0.05).And after model,the natural group,sham-operated group, pre-needle group,pre-moxibustion group,pre-acupuncture and moxibustion group,the average escape latency of each group there was no significant difference between themselives(P>0.05).But after model,the average escape latency of the natural group,sham-operated group,pre-acupuncture group, pre-moxibustion group,pre-acupuncture and moxibustion group were significantly less than after the model of the model group, there is a very significant difference(P<0.01). 2 Influence of pre-stimulation on Alzheimer rat models' the number of errors entered into blind-side in channel-type water maze.Analysis fromadifferent time period,former model, the average the number of errors entered into blind-side of each group was no significant difference(P>0.05).After model,the average escape latency the number of errors entered into blind-side significantly increased than that of former model,there is a very significant difference(P<0.01).And after model,the natural group,sham-operated group, pre-needle group,pre-moxibustion group,pre-acupuncture and moxibustion group,the average the number of errors entered into blind-side of each group comparing with former model each group itself,there was no significant difference(P>0.05). And after model,the natural group,sham-operated group, pre-needle group,pre-moxibustion group,pre-acupuncture and moxibustion group,the average the number of errors entered into blind-side of each group there was no significant difference between themselives(P>0.05).But after model,the average the number of errors entered into blind-side of the natural group,sham-operated group,pre-acupuncture group, pre-moxibustion group,pre-acupuncture and moxibustion group was significantly less than the model group after the model, there was a very significant difference(P<0.01).3 After model,the natural group,sham-operated group, pre-needle group,pre-moxibustion group,pre-acupuncture and moxibustion group has the remarkable enhancement in activity of SOD in rats' brain hippocampal tissue compared to the model group,there is significant difference(P<0.01),and the activity of SOD in rats' brain hippocampal tissue among the natural group,sham-operated group,pre-needle group, pre-moxibust ion group,pre-acupuncture and moxibustion group, there was no significant difference(P>0.05);the natural group,sham-operated group,pre-needle group,pre-moxibustion group,pre-acupuncture and moxibustion group has obviously reduced in activity of NOS in rats' brain hippocampal tissue compared to the model group,there is significant difference (P<0.01),and the activity of NOS in rats' brain hippocampal tissue among the natural group,sham-operated group, pre-needle group,pre-moxibustion group,pre-acupuncture and moxibustion group,there was no significant difference(P>0.05).Conclusionpre-stimulation with The acupuncture and moxibustion has the function to improve the SOD activity,depress the activity of NOS in rat,hippocampal area,and improve learning and memory ability of AD rats. |
