| ObjectiveAlcohol is the major constituents of hard drink,and also one of the main reasons to cause traffic accident drive when intoxicated or drunk drive is based on the blood alcohol concerntration(BAC) of the road traffic safety act.But when the driver losses blood caused by hurt and other reasons,policeman couldn't detect the blood alcohol concerntration.At this time,legal medical expert may deduce blood alcohol concerntration by body fluids and tissues,such as vitreous humor,gall,ncurolymph, marrow,muscle and so on.We adopt headspace gas chromatography to detect the alcohol concentration of boold,vitreous humor,and muscle in rats with alcohol,and establish a method for deducing the relationship between the alcohol in the blood, vitreous humor and the muscle of rat.MethodAdopt headspace gas chromatograpgy to detect the alcohol concentration in the blood,vitreous humor and the muscle of rats with alcohol after 1,2,3,4,5 hours.ResultsAfter the intragastric administration with ethanol,the rats were killed at 1h,2h,3h, 4h,5h,the blood,vitreous humor and muscle were get and analyzed by linearity regression.The linear equation between time(1h-5h) and the concentration of ethanol in blood is y=226.3402-35.6091x(r~2 =0.9798,r =0.9898),p＜0.005.The linear equation between time and the concentration of ethanol in the vitreous humor is y=229.3067-30.0944x(r~2=0.9794,r=-0.9896),p＜0.005.The linear equation between time and the concentration of ethanol in the muscle is y=181.3285-21.7791x (r~2 =0.9895,r =-0.9947),p＜0.005. DiscussionIn the present study,after rats were fed with ethanol,the 3 and 4 hours'BAC/ VAC were 0.85 and 0.79,which fully demonstrated that the distribution of ethanol had reached a balance in the rat body in 3 hours(absorption of the latter).The BAC/VAC, BAC/MAC of the 2 hours group were higher than the theoretical value,which shows that the distribution of ethanol had not yet reached a balance at this time(in the absorption period).After the rats were fed with ethanol,the 1,2,3,4,5,8,10,12-hour's BAC/VAC,BAC/MAC of the experimental group showed that:BAC/VAC,BAC/ MAC values decreased gradually with time,and its elimination rate was slower than the elimination rate of BAC.The result of the linear correlation and regression statistical analysis for the average concentration of ethanol in Blood,vitreous humor and muscle was positive,the correlation is highly significant(p＜0.005).The result showed that the method for deducing the relationship between the alcohol concentration in the blood,vitreous humor and muscle of the rats is reasonable.ConclusionTo establish a method for deducing the relationship between the alcohol concentration in the blood,vitreous humor and muscle of the rats with alcohol from 1h to 5h,BAC =-44.9045+0.5948VAC,BAC=-66.73239+1.60569MAC,this provides a reference frame for deducing BAC. |
