Objective:To investigate the influence of hypoxia on the ACE2 gene expression of cultured rats' vascular smooth muscle cellsMethods:The thoracic aorta was obtained from Sprague-Dawley rats under careful sterile conditions.The smooth muscle cell(SMC) were isolated from the media of the vessel and use attachment-block culture.Use a self-made hypoxia equipment,detect it's hypoxia effect and found a plan of hypoxia in vitro.To observe the cytoactive When cells were cultured under hypoxia conditions,the general morphology change was observed,cell counting was used to investigate the cytoactive.Total RNA was purified from tissue with TRIZOL reagent.Semi-quantified RT PCR was used to investigate the ACE2 gene expression in hypoxia SMC,α-Actin as control gene.Results:The rats' vessel smooth muscle cell line was successfully obtained and cultured in vitro.Self-made hypoxia equipment can meet the experiment requirement Hypoxia doesn't induce an obvious change in cytoactive.Nevertheless,the expression of ACE2 is decreased after 8 hours culturing in hypoxia,and significantly increase in longer hypoxia groups.After hypoxia 24h to recover oxygen the expression of ACE2 is decreased,after recover oxygen 8h the expression of ACE2 return normal.Conclusion:Hypoxia and after hypoxia to recover oxygen have an effect on the expression of ACE2.The expression of ACE2 is decreased after 8 hours culturing in hypoxia,and significantly increase in longer hypoxia groups.After hypoxia 24h to recover oxygen the expression of ACE2 is decreased,after recover oxygen 8h the expression of ACE2 return normal.
