Objective:Sepsis is an important disease which threaten human health.As a later period cytokine,High mobility group protein B1 (HMGB1) is a key factor which lead to killing effect suffering from sepsis.Observing the expression and significance of several cytokine including HMGB1 caused by using penehyclidine hydrochloride treat rat with Sepsis -associated lung injury.To investigate the efficacy of penehyclidine hydrochloride in rat with Sepsis-associated lung injury.Methods:In a sepsis model by cecal ligation and puncture,ninety SD rats were randomly devided into sham operation,acute lung injury,and penehyclidine hydrochloride treatment(0.45mg/ kg i.m) groups.After operated 24,48,72 hours,the concentration of HMGB 1 and Macrophage migration Inhibitory Factor(MIF) in the serum were measured;wet to dry weight ratio(W/D) and the concentration of the Aquaporin-5(AQP-5) in the bronchoalveo-Lar lavage fluid(BALF) were measured,the lungs of each group were sampled for light microscope.Results:Contrasting with sepsis-associated lung injury group,the concentration of HMGB1,MIF in the serum and W/D in the penehyclidine hydrochloride treatment group after operated 24,48,72 hours decreased significantly(P<0.01);the concentration of AQP—5 in the BALF in the penehyclidine hydrochloride treatment group at corresponding period increased significantly(P<0.01).Conclusion:penehyclidine hydrochloride inhibits the expression of HMGB1 and MIF in serum and promote the expression of AQP-5 in BALF,had effects on acute lung injury induced by sepsis.
