| Objective:To further clarify the major environmental risk factors in the occurrence of gastric cancer,population-based case-control study on molecular epidemiology was carried out in Yixing Jiangsu Province, where has the high incidence of gastric cancer.To study and research the relationship between RUNX3 gene mutation and the occurrence of gastric cancer in China,three polymorphic sites with functional significance of RUNX3 gene were screened.Based on this,the environment-gene interactions will be evaluated,and in the end the mechanism in the molecular genetics which lead to the incidence of gastric cancer will be clarified.Furthermore,this study will guide the prevention and therapy for high-risk groups.Methods:The living material of 312 gastric cancer patients and 329 control were collected in Yixing City,Jiangsu Province.The general state of health was compared under SAS software,to find out the relationship between the occurrence of gastric cancer with age,sex and smoking,alcohol consumption etc.At the same time,The blood was drawed in the case with the control group.The DNA was got out to study 3 SNPs of the RUNX3 gene rs6672420 T＞A,rs11249208 A＞G, rs11249206 T＞C in the case with the control group through the way of PCR-RFLP. Results:1.male cases of gastric cancer cases are most but no significant difference.however,we also found femal cases were more in the advanced stage and.the result was obvious.The incidence of gastric cancer increases with age,especially after the age of 45.Gastric cancer cases have more bad habits such as smoking more,drinking etc.2.RUNX3 gene SNP rs11249206 T＞C has significantly difference in gastric cancer group and control group.Conclusion:1.The incidence of gastric cancer is a multi-factor,stage.There are many known factors involved in the occurrence of gastric cancer.2.RUNX3 gene is an important tumor suppressor gene in gastric cancer,the mutation of RUNX3 gene is a very important factor to lead gastric cancer.
