Objective:To analyze the relations between the frequencies of IL-2R and IL-7R gene polymorphisms and the susceptibility to idiopathic demyelinating optic neuritis.Methods:1.subjects:a case-control research was performed on 72 IDON patients (37male and 35 female;mean age,29 years) and 81 healthy individuals as control group(48male and 33 female;mean age,34 years).All patients fulfilled the criteria of IDON.Control subjects from the PLA hospital consisted of unrelated people who reported themselves free of chronic inflammatory disease and no personal or familial history of autoimmune disease.2.methods:DNA was extracted from peripheral blood leukocytes;PCR was used to amplify the aim genetic sequence.;the gene polymorphisms of IL-2R were analyzed by restriction fragment length polymorphisms (RFLP) assay.;and the gene polymorphisms of IL-7R were analyzed by gene sequencing.3.Statistical analysis:the clinical data such as the age wae expressed as Mean.Hardy-Weinberg equilibrium(HWE) was tested to ensure that control samples were within allelic population equilibrium.The x~2 test was used to compare genotype and allele frequencies between the IDON and control populations.Or was calculated using the approximation of Woolf for the 95%confidence interval(c.i.).A P-value of less than 0.05 was considered statistically significant.All statistical analyses were performed using SPSS11.5 statistical analysis software.Results:1.No significant difference of genotype and allele frequencise of IL-2R existed in IDON group compared with the control group(x~2=0.4102,P =0.8146;x~2=0.4134,P=0.5202,genotype and allele frequency respectively).2.No significant difference of genotype frequency of IL-7R existed in IDON group compared with the control group(x~2=3.787,P=0.15);while the frequencise of IL-7R allele increased significantly in IDON group compared with those of healthy controls(x~2=4.743,P=0.0294 ),C allele may contribute to the increased susceptibility of IDON(OR=1.95,95% C.I.1.07-3.55).Conclusion:1.The frequencies of IL-2R AA genotype and A allele higher in IDON group compared with those of healthy controls,but no significant difference existed.2.The frequencies of IL-7R C allele increased significantly in IDON group compared with those of healthy controls,the SNP of IL-7R gene may contribute to the risk of IDON with odds ratios 1.95.
