It is well known that chemotherapy is an improtant method in combined therapy of tumor,it plays a realistic role in the short-term and prospective efficacy.But the two shortcomings of multi-drug resistance and leipo-target mostly lower the effect of the chemotherapy in clinical application.The over-expression of P-gp has been associated with the development of multi-drug resistance(MDR) in cancer cells.For these reasons,in this study,doxorubicin content alginate nanoparticle(DAN) modified with transferrin(TDAN) was obtained by a reverse phase microemulsion process.The nanoparticle with the sizes of 100-300nm were prepared by heptane as organic dispersed medium,span-80 as surfactart,DOX and ALG as raw materials and the transferrin as specific targeting agent to cancer cells was conjugated to the DAN by an EDC cross linking reaction.The effects of preparation conditions such as the concentrations of ALG and surfactant, antibody dosage and reaction time of antibody with microspheres on TDAN properties,were investigated by transmission electron microscopy,laser light scattering detector,atomic adsorption spectroscopy,and flow cytometer for the immunological activity of antibody on TDAN.Results showed that the spherical TDAN had an average size of about 170nm,and most size distributions between 100nm and 300nm.The Tf content in the nanoparticle was 42.3%,and the drug loading was 11.9%,and released the drug in an effect concentration for a long time.The Tf on the surface of TDAN was active and could guide the particles to human liver tumor cells.It was concluded that as a new carrier of DOX,TDAN could be used to effectively deliver DOX to the tumor by guidance of its Tf receptor-targeted in an effective therapy concentration of DOX and less side effects on the heart and the kidney.In summary,this study indicated that the Tf receptor-targeted and doxorubicin-loaded nanoparticle,as an carrier of anticancer drug,was hopeful to achieve clinical cancer treatment.
