Part 1 Effect of Erigeron breviscapus on renal function, histology and expression of p27 in diabetic rats.Objective To investigate the effect of Erigeronbreviscapine on the renal function structure,histology and the expression of p27 in diabetic rats.Methods 85 normal male rats were selected.Rats were randomly divided into three groups:control group(C),diabetic group(D),and Erigeronbreviscapine intervention group(EB).D and EB groups were induced by injecting streptozotocin(STZ) to become the diabetic model.Later Erigeronbreviscapine was injected into rats cavity in EB group by 20mg.kg-1d-1 dose.Normal Saline was injected into rats cavity in C and D groups by the same dose.After two weeks 10 in C group,10 in D group,and 12 in E group were randomly selected and killed.At six weeks all rats were killed.Then blood specimen of all rats were collected to examine blood glucose(Glu),serum creatinine(Cre),blood urea nitrogen(Bun).The changes of the mean glomerular area (MGPA) and the average glomerular volume(MVG) of the three groups were observed by histological.We detected the expression of p27 by immunohistochemical.Results①Comparing with C group,the weight of D group and EB group were increased;Comparison of the level of Cre and Bun:D group>EB group>C group. Compare with D group,the level of Cre and Bun were lower.Cre decrease was more obvious in 2 weeks(P<0.01).②Comparing with C group,the renal index,MGPA and MVG of both diabetic group and breviscapine intervention group were increased.It is likeDgroup>EB group>C group.There are significant differences(P<0.05).③The expression of p27 is D group>EBgroup>C group.(P<0.05) and 6w group is higher than 2w.Conclusion Cell hypertrophy and p27 protein expression are increased significantly in the early stage of diabetic nephropathy,and those can improve by the Erigeron breviscapine intervention. Part 2 Effect of Erigeron breviscapus on renal apoptosis and expression of gene Akt,p70s6k and 4E-BP-1 in diabetes ratsObjective To investigate the effect of Erigeron breviscapine on apoptosis and the expression of Akt,P70S6K and 4E-BP-ⅠmRNA in kidney of diabetic rats.Methods 85 normal male rats were selected.Rats were randomly divided into three groups:control group(C),diabetic group(D),and Erigeronbreviscapine intervention group(EB).D and EB groups were induced by injecting streptozotocin(STZ) to become the diabetic model.After that Erigeronbreviscapine was injected into rats cavity in EB group by 20mg.kg-1d-1 dose.Normal Saline was injected into rats cavity in C and D groups by the same dose.After two weeks 10 in C group,10 in D group,and 12 in E group were randomly selected and killed.At six weeks all were killed.Apoptosis were identified by terminal deoxy nucleotidyl transferase mediated d-UTP nick end labeling(TUNEL).Expression of renal Akt,p70s6k and 4E-BP-1 mRNA were detected by real-time quantitative with SYBR GreenⅠ.The above different indicators were Compared.Results①Compare with C group,the apoptosis cell in tubules of D group and EB goup are increase obviously at 2 and 6 weeks,D group higher than EB group.Apoptosis in glomeruli is not detected at 2 weeks,is detected rarelly at 6 weeks.②The expression of Akt mRNA are increased at both 2 weeks and 6 weeks in D group.At 2 weeks it showed D group>C group>EB group and there is no significant difference,At 6 weeks it manifested as EB group>C group>D group and there are significant differences.Compare with C group,D group is higher but EB group is highest.There is no significant difference in the expression of p70s6k mRNA and 4E-BP-ⅠmRNA in each group.Conclusion Both apoptosis and Akt mRNA expression are increased.Erigeron breviscapus could inhibit renal apoptosis in diabetic rats and Erigeron breviscapus could improve the expression of Akt mRNA as the times go.PI3K/Akt signaling pathway may be involved apoptosis in diabetic nephropathy which can be protected by Erigeron breviscapine.
