【Objective】Bone marrow-derived mesenchymal stem cells(MSC) markersed with green fluorescent protein(GFP) were tansplanted to the multiple organ dysfunction syndrome(MODS) in rabbits.We assayed the effect of MSC's transplantation on rabbit with MODS and the distribution in the host,in order to d provide a new way for the treatment of MODS.【Methods】Extected bone marrow from the rabbit Under sterile conditions,Isolated and expanded MSC,markers MSC with GFP by lentivirus.30 rabbits were randomly divided into three groups:normal group,model group,treatment group.The model group,the treatment group were used to establish MODS rabbits by hemorrhagic shock combined with endotoxin shock.We injected the model with MSC by 3×106cells/kg in ear margin vein after successfully modeled for 24 hours,then tested the changes of PaO2,PaCO2,BUN,ALT,CK-MB of rabbit in the model and treatment group on the first day,the seventh day,the fourteenth day and the twenty-eighth day.On the twenty-eighth day,kidney,lung,liver,brain and heart tissue of the last two groups were taken to make pathological sections and extract DNA,then detected the distribution of GFP gene by PCR in these organs.【Results】we acquired MSC which can grow adherently,and have the ability to differentiate into osteoblasts and adipogenic cells,and still maintain the ability of differention after GFP markers by lentivirus.Exterior antibody expressed CD34-,CD45-,CD73+,CD105+ that conformed to the characteristics of MSC.Compared to control group the physical index PaO2,PaCO2,BUN,ALT,CKMB were significantly different,and similar to clinical case. Compared with model group,PaO2 of rabbit in treatment group detected on the seventh day was increase,which mean function of lung has improved;BUN of rabbit in treatment group detected on the seventh day was decrease,which mean function of renal has improved;ALT of rabbit in treatment group detected on the fourteenth day was decrease,which mean function of liver has improved,There was no difference in other more.There was no difference in mortality,too.The GFP-MSC could be found on the section of lung,kidney,liver,brain,but specific differentiated cell type was not detected.Meanwhile By PCR technology the GFP gene was found in the tissues mentioned above,And the distribution is highest in the lung tissue,followed by liver,kidney,brain and heart.【Conclusion】(1) Established in vitro of MSC mark GFP technology such as isolation, culture,identification of directional differentiation and so on.(2) Successfully established rabbit MODS model.(3) MSC that tranplated by vein infusion can integrate into lung,kidney, liver and brain.And the distribution is highest in the lung tissue,followed by liver,kidney, brain and heart.Compared with model group,PaO2 of rabbit in treatment group detected on the seventh day was increase,which mean function of lung has improved;BUN of rabbit in treatment group detected on the seventh day was decrease,which mean function of renal has improved;ALT of rabbit in treatment group detected on the fourteenth day was decrease, which mean function of liver has improved.The mechanism of MSC work are by inhibitting excessive inflammatory immune response and adjusting system balance;produce cytokines to regulate the network balance;involved in damage repairment and this needs us to make further study.
