| Objective:The experiment mainly study the male Sprague-Dawley rats. To study the influence of treadmill exercise on the utrastructure,the content of the hydroxyproline and the dynamic variation of IGF-I expression in tendon,we have a comprehensive study on the organizational structure,biochemistry, as well as growth factor.Through the above study,we can investigate the extracellular matrix of tendon and the dynamic variation of IGF-I expression to exercise through the response to a bout of treadmill exercise and long-term treadmill exercise for 12 weeks.Methods:â‘ A bout of treadmill exercise group:30 male Sprague-Dawley rats were divided into 2 groups randomly,including 15 rats selected as control group and 15 rats as a bout of exercise group.â‘¡Long-term treadmill exercise group:30 male rats were used in this model,including 15 rats selected as control group and 15 rats as long-term exercise group,and the experimental groups were subjected to running exercise for 12 weeks.Parameters and methods of measure:The structure of fiber,fibril and activity of tenocytes were observed with optics microscope and electronic microscope.The concentration of the Hydroxyproline were measured by kits test methods.The concentrations of IGF-I of achilles tendon and IGF-I in serum were measured by Elisa methods.The expression of procollagen IGF-I in mRNA level were analyzed by real-time quantitative PCR technique.Results:(1)The effect of a bout of treadmill exercise on the Achlles tendon of the rats:â‘ Morphology observation showed the interval increase and disarrangement of collagen fiber of the achilles tendon after a bout of treadmill exercise. At the same time,The tenocytes become active.â‘¡The mass of the tendon and the content of the Hydroxyproline did not significantly increased compared with the control group.â‘¢The concentrations of IGF-I mRNA and protein in achilles tendon and serum were not significantly increased compared with the control group.(2)The effects of long-term treadmill on the Achlles tendon of the rats:â‘ Rphology observation showed the arrangement and rugular of the collagen fiber of the rats. The tenocytes metabolism became active.â‘¡Although the mass of the tendon and the content of the Hydroxyproline increased,but there were not significantly increased compared with the control group.â‘¢The concentrations of IGF-I mRNA and protein in achilles tendon and serum were significantly increased compared with the control group(p<0.01).Conclusion:â‘ After the bout of treadmill exercise, Morphology observation show the interval increase and disarrangement of collagen fiber of the achilles tendon,However, Rphology observation show the arrangement and rugular of the collagen fiber of the rats.The tenocytes metabolism become active as the long-term treadmill exercise.The research show thatthe ultrastructural changes of the tendon is temporary after the bout of treadmill exercise,However,There is a long-term adaptive changes in the ultrastructure of tendon by the long-term treadmill exercise.â‘¡The mass of the tendon and the content of the Hydroxyproline do not significantly increase compared with the control group.Although the mass of the tendon and the content of the Hydroxyproline increase,but there are not significantly increase compared with the control group.â‘¢The concentrations of IGF-I mRNA and protein in achilles tendon and serum are not significantly increased compared with the control group.The concentrations of IGF-I mRNA and protein in achilles tendon and serum are significantly increased compared with the control group(p<0.01).The study shows that IGF-I in the serum and local of the tendon can cooperates on mediating the metabolism of tendon. |
