Objective To study the significance of iNOS and VEGF in acute liver failure(ALF), by detecting the expression of them in liver rats with ALF, observing the effect of Aminoguanidine, and analyzing the correlation between iNOS and VEGF.Methods Sixty rats were randomly divided into six groups, ten rats each group. The NS(normal saline) control group and AG(Aminoguanidine) control group were injected intraperitoneally with NS and AG (100mg/Kg), respectively. To induce acute liver failure, rats were injected intraperitoneally with D-GalN(400mg/Kg) and LPS(50μg/Kg), dissolved in NS(in the volume of 1ml). ALF model group were devided into D-6 group and D-12 group, and the rats were executed atfter 6h, 12h, respectively. AG intervention groups were devided into A-6 group and A-12 group, which were administered D-GlaN and LPS, 30 minutes after being injected intraperitoneally with AG (100mg/Kg i.p); the rats were executed after 6h, 12h respectively. Blood was taken from portal vein to detect liver function.The liver tissue specimens were taken to detect Nitric Oxide Concentration which was measured by nitrate reductase method. The expression of iNOS and VEGF in liver tissue of rats was evaluated by immunohistochemistry assay. Image analysis system was used to analyze the average gray value.Results (1)The concentration of NO in NS group and AG group was 14.39±2.99 umol/L, 13.39±2.83 umol/L,respectively, t=0.77,Ρ=0.4527. The levels of NO in AG intervention groups significantly decreased compared to ALF model groups, which were 15.79±2.53 umol/L, 26.61±7.92 umol/L at 6h, repectively, t=3.09,Ρ=0.0109, and 30.83±5.08umol/L, 48.38±4.18umol/L at 12h, respectively, t=7.70,Ρ<0.001. (2)There were no stained iNOS and VEGF immunoreactive cells in the control groups. The experimental groups highly expressed iNOS and VEGF. The mean gray value of iNOS and VEGF in AG intervention significantly increased compared to ALF model groups, which were 119.18±6.55/135.08±5.37, 81.02±5.86/124.33±3.80, respectively, at 6h(P<0.05); and 151.52±5.87/155.34±5.82, 130.71±7.16/145.69±3.35, respectively, at 12h (P<0.05). (3)Spearman correlation showed that iNOS was significantly correlated with VEGF, and the spearman correlation coefficient between them was 0.8859, (P<0.05).Conclusions (1)The concentration of NO elevated in the liver tissue of rats with ALF, which was inhibited by Aminoguanidine. (2) High expression of iNOS and VEGF was observed in liver tissue of rats with ALF, and Aminoguanidine has an suppressive effect on them. (3)INOS showed positive correlation with VEGF. (4)These results suggest that iNOS and VEGF might play important roles on neovascularization, reconstruction of hepatic sinusoid and hepatocyte regeneration.
