Objective The purpose of the present study is to detect the expression of RhoC and LyGDI, analyze the correlation between RhoC and RhoGDI and evaluate clinical significance of them on non-small cell lung cancer.Methods We collected samples on lung cancer tissue with corresponding paracancerous tissue by operation in the department of cardiothoracic of the First Affiliated Hospital of Soochow University from July 2007 to October 2008, each of tissue have forty - eight samples and cancer tissue were confirmed to be primary non-small cell lung cancer by postoperative pathological examination.Western Blot and Immunohistochemistry were used respectively to detect RhoC and LyGDI in cancer tissue and corresponding paracancerous tissue, and analyzed statistically with clinical data.Results 1.The expression levels of RhoC and LyGDI on non-small cell lung cancer tissue and corresponding paracancerous tissue:(1)Immunohistochemistry suggested that the positive signal of RhoC and LyGDI were all fixed in the cytoplasm in cancer tissue and corresponding paracancerous tissue;the positive rate of RhoC and LyGDI were respectively about 77.1% and 68.8% in cancer tissue;both of them have only a small amount of expression in paracancerous tissue.(2)Western Blot suggested that RhoC and LyGDI all have expressed in cancer tissue and the corresponding paracancerous tissue,the molecular weight of them are respectively 23kDa (Dalton),27kDa;the relative expression of them are respectively (2.197±0.220,1.035±0.118),(1.810±0.099,0.859±0.091)in cancer tissue and the corresponding paracancerous tissue, the expression levels of RhoC and LyGDI were more higher in cancer tissue than them in the corresponding paracancerous tissue(P=0.001,P=0.002).2.The relationship between expression of RhoC and LyGDI on non-small cell lung cancer:We examined the gray value of the bands by Western blot with Spearman correlation analysis and found that RhoC and LyGDI had no relationship (rs=-0.011,P=0.942).3.The relationship between the expression of RhoC and LyGDI and clinical feature:We did Chi-square on the relationship cinical feature and expression of RhoC and LyGDI on non-small cell lung cancer which tested by Immunohistochemistry, and the result suggest:(1)The positive rate on RhoC was correlated TNM stage(P=0.022) or lymph node metastasis(P=0.011), but RhoC expression was not in association with sex(P=0.247),age(P=0.616),differentiation(P=0.428)or histology(P=0.897).(2)The positive rate on LyGDI was correlated with TNM stage(P=0.031) or lymph node metastasis (P=0.011),but LyGDI expression was not in association with sex (P=0.834),age (P=0.613) , differentiation (P=0.654) or histology (P=0.724).Conclusion 1. The expression levels of RhoC and LyGDI were more higher in cancer tissue than them in the corresponding paracancerous tissue, indicated that they played a role in the occurrence of non-small cell lung cancer.2.The positive rate in RhoC was correlated with the increasing of TNM stage or lymph node metastasis,indicated that it could promote the disease progression and lymph node metastasis of non-small cell lung cancer.The positive rate in LyGDI was correlated with the of TNM stage or lymph node metastasis,indicated that it could inhibit the disease progression and lymph node metastasis of non-small cell lung cancer.3.The expression levels of RhoC and LyGDI had no relationship.
