[Objective]: To investigate the role of JAK2/STAT3 in the expression of MMP-10 in Human lung adenocarcioma cell line and explore the molecular regulatory mechanism of MMP-10 expression.[Methods]: A549 cells were cultured in RPMI 1640 medium containing 10% fetal bovine serum for 24h in free serum medium after 50-70% cells pasted on the wall. 0 ng/ml, 12.5 ng/ml, 25 ng/ml, 50 ng/ml IL-6 were given to A549 cells, Meanwhile the cells were pre-incubated with 50μM AG490 for 4 h and then were stimulated respectively determined by 25 ng/ml IL-6 for 24h. The expression of JAK2 mRNA, STAT3 mRNA and MMP-10 mRNA was detected with Real time RT-PCR assay. The expression of MMP-10 protein was assessed by Western blot.[Results]: 1.In A549 cells, STAT3 mRNA levels at 25ng/ml IL-6 was higher(43.87%) than at 0ng/ml IL-6(P<0.05); 50μM AG490, a specific JAK2 inhibitor, together with 25ng/ml IL-6 decreased STAT3 mRNA expression by 36.73%, compared with 25ng/ml IL-6 (P<0.01). On the contrary, MMP-10 mRNA levels in A549 cells pre-incubated with 50μM AG490 were significantly higher than with 25ng/ml IL-6 (P<0.01). 2.Compared with 0 ng/ml IL-6, MMP-10 protein levels at 12.5, 25, 50ng/ml Il-6 were higher (P<0.05) with a peak expression at the concentration of 25 ng/ml IL-6 (P<0.01); 50μM AG490, together with 25ng/ml IL-6 reduced MMP-10 protein expression compared with 25ng/ml IL-6 (P<0.05).[Conclusion]: It is suggested that the JAK2/STAT3 pathway might be involved in the expression of MMP-10 in A549 cells.
