IFN-γis one of common infectious factors and the main component of inflammation.During the pregnancy,the pregnant women are more vulnerable to various infectious factors,which may have harmful effect on the central neural system of fetus.In the present article,we stimulated neural stem cells of embryonic D16 by IFN-γ(200 U/ml)in vitro to investigate the IFN-γeffect on embryonic stem cell.In this study, we successfully established the method of embryonic stem cell culture in vitro,by which more than 90%purity of embryonic stem cells was obtained. The embryonic stem cell is able to self-renew and multi-differentiate to neurons,oligodendrocytes and astrocytes.As a infectious factor,IFN-γ, in the high dose,has the toxic effect on embryonic stem cell.IFN-γcan up-regulate the expression of MHCⅠand MHCⅡmolecules and increase the differentiation of neuron and oligodendrocyte and decrease the astrocytes differentiation.All the results indicated that the severe inflammatory response will probably change the condition of neural stem cell growth and differentiation in fetus early development so as to lead to dysfunction post-birth.
