| ObjectionTo establish model of rabbit with hypercholesterolemia-atherosclerasis and observe blood-fat and lipoprotein-associated phospholipase A2 concentration in the serum. We will investigate the relationship between lipoprotein-associated phospholipase A2 and atherosclerasis and in the influence of simvastatin to lipoprotein-associated phospholipase A2 in rabbits.MethodsFifty-six healthy male New-Zealand albion rabbits were devided into three groups: normal control , high cholesteroldiet,simvastatin. After feeding for the forth, eighth,twelfth weeks,we will collect the empty stomach blood sample of these rabbits through central artery at awake state (fasting about twelve hours),centrifugalization (3000rpm, 10min),blood serum was stored at -20℃.We will evaluate the concentration of total cholestrrol (TC), triglyceride (TG), low density lipoprotein (LDL),high density lipoprotein (HDL) and lipoprotein-associated phospholipase A2 (LP-PLA2) by ELISA. And to observe the pathomorphology of the rabbits abdominal aorta and thoracic aorta by the optical microscope.ResultsDuing the experiment, we found:1. The concertration of blood-fat and LP-PLA2 in the normal control is no significant differences at the forth, eighth, twelfth weeks.2. After feeding for the forth, eighth, twelfth weeks, the comparison of high cholesteroldiet group and normal control group, the concentration of the TC, LDL, LP-PLA2 in the former are increased significently compared to the latter (p<0.01), the concentration of HDL in the former is increased compared to the latter (p<0.05), but had a descendent trend. 3. After feeding for the eighth weeks, the comparison of high cholesteroldiet group and normal control group, the concentration of TG in the former is increased significantly compared to the latter (p<0.01).4. Simvastatin group: at the end of the eighth, the comparison of high cholesteroldiet group and normal control group,, the concentration of LP-PLA2 in the former is no significant differences compared to the latter (p<0.05); at the end of twelfth weeks, p is less than 0.01.Conclusions1. LP-PLA2 plays a role in the course of atherosclerasis.2. Simvastatin has a significant effect in the term of decreasing the concentration of LP-PLA2. |
