Expression And Significance Of P63 In The Course Of Cornea Healing After Injury In Rats

PURPOSE:Scraped the rats' corneal epithelial to injure the cornea and start the course of corneal healing,and detect the different expression of p63 at different times.To research the significance of p63 in the course of rats' corneal epithelial healing.METHODS:1.Executed rats in 0,6,12,24,48hours after their apart of corneal epithelial had been scraped.2.Fluorescein sodium staining was used to observe the course of healing through slit lamp.3.Paraffin sections was made after the rats been sacrificed,the expression of p63 was detected by immunofluorescence reaction and analyze the different expression of p63 at different times in central cornea and corneal limbus.4.RNA was extracted after the rats been executed and the expression of p63 mRNA was detected by RT-PCR, and analyze the different expression of p63 mRNA at different times and different locations.RESULTS:1.the result of immunofluorescence reaction:p63 only express in corneal limbus rather than central cornea,p63 expression was negative in the group of central cornea and there were fundamental difference between them and any groups in corneal limbus.The expression of p63 was increased along with the course of corneal healing,and arrived the peak around 24th hour after cornea injury.There was no statistical significance between the average absorbance of normal group and Oh group(P＞0.05 ),or between 6h group and 48h group(P＞0.05 ) But there were high statistical significance between any other two groups(P＜0.01). 2.The result of RT-PCR:The expression of p63 mRNA increased along with the course of corneal healing,and arrived the peak around 24th hour after cornea injury.The expression of p63 mRNA in central cornea were 0 and there were high statistical significance between them and any groups in corneal limbus(P＜0.01). There was no statistical significance between the average absorbance of normal group and Oh group(P＞0.05 ).There were statistical significances between normal group and Oh group,Oh group and 6h group,12h group and 48h group.And there were high statistical significance between any other two groups(P＜0.01).3.The injured corneal healed in 24hours.CONCLUSIONS:1.In normal cornea and in the course of corneal healing, p63 only express in the area of corneal limbus,rather than central cornea.2.The expression of p63 was closely related with the course of corneal epithelial healing and the proliferation of cornea limbal stem cells,p63 probably can promote the healing of cornea.
