Background:As the major component of turmeric,curcumin has a variety of pharmacological effects.Studies have shown that curcumin could inhibit the growth of some tumor cells including prostate cancer cells,and prevent liver and kidney injury induced by chemotherapy or radiotherapy.The main therapy of androgen-independent prostate cancer is chemotherapy or radiotherapy.It was hypothesized that the combination effects of curcumin with chemotherapy not only can inhibit the growth of prostate cancer cells,but also can protect liver and kidney, which appears to be effective complex treatment for prostate cancer.Objective:This study aimed to confirm the combination effects of curcumin with paclitaxel on androgen-independent prostate cancer PC3 cells and PC3 transplanted tumors in nude mice,especially in proliferation,differentiation and invasion ability of PC3.To find effective complex treatment measures for prostate cancer.Methods:The cell viability,senescence and invasive capability of PC3 were detected respectively by CCK-8,β-galactosidase staining,scratched invasion assay.The mRNA relative quantity of Id1,PCNA and MMP2 was observed by SYBR GreenⅠbased real-time RT-PCR in PC3 cells and PC3 transplanted tumors in nude mice.Results:Curcumin combined with paclitaxel synergistically decreased cell proliferation of prostate cancer PC3 cells(p<0.01).Scratch experiments showed that the invasive capability of PC3 cells could also be reduced by the combination of curcumin with paclitaxel.Low concentration of curcumin(5μmol/l) can induce cell senescence(p<0.05).In order to confirm the combination effects of curcumin with paclitaxel,the mRNA expression of Id1,PCNA and MMP2 were detected by Real time RT-PCR in PC3 cell and tumors transplanted with PC3 cells in nude mice.The results showed that mRNA expression of Id1,PCNA and MMP2 were reduced obviously in PC3 cells and transplanted tumors(p<0.01).The study confirmed that the combination of curcumin with paclitaxel could reduce cell proliferation, differentiation and invasive capability of PC3 in vivo and in vitro.Conclusions:1.Curcumin can inhibit cell proliferation,invasive capability and induce cell senescence in PC3.2.Curcumin can decrease the mRNA expression of Id1,PCNA and MMP2 in PC3 cells and tumors transplanted with PC3 in nude mice.3.Curcumin combined with paclitaxel can synergistically decrease cell proliferation and invasive capability;decrease the mRNA expression of Id1,PCNA and MMP2 in PC3 cells.4.Curcumin combined with paclitaxel also have synergistically effects in vivo as same as in PC3 cells.
