The Effects Of Ge Pi Decoction On Experimental Rats With Type 2 Diabetic Mellitus

Objective:To investigating the therapeutic effect of Ge Pi Decoctiont on rats with type 2 diabetic and its mechanism for clinical application.Methods:Type 2 diabetic rat models induced by STZ were randomly divided into diabetes model group(n=11),Metformin group(n=15),Ge Pi Decoctiont group(n=16).And set up the control group(n=10).General situation were observed during and after treatment.Blood glucose,total cholesterol(TC),triglyceride(TG),blood insulin,24h urinary albumin,monocyte chemoattractant protein-1(MCP-1),8-iso-prostaglandin F2a(8-iso-PGF2α),tumor necrosis factor -α(TNF-α),IL-6 were detected.Kidney weight was measured.And calculated insulin sensitivity index(ISI) and kidney weight index.The pathological changes of the rats pancreas and kidney were observed in light microscope.Results:The model rats established by injection STZ after feeding high glucose and lipids conformed to type 2 Diabetes rats.Ge Pi Decoction can improved the general conditions of diabetic rats.Compared with the simple model group,blood glucose,TC,TG,fasting insulin,24h urinary albumin,kidney weight and kidney weight index of the Ge Pi Decoction group decreased,ISI improved.The changes of blood glucose in Ge Pi Decoction group were weaker than Metformin group.There was no significant difference between two treatment group in TC,TG,insulin drop.The changes of 24h urinary albumin and ISI in Ge Pi Decoction group were greater than Metformin group. MCP-1,8-iso-PGF2α,TNF-α,IL-6 of Ge Pi Decoction group were decreased than the simple model group.There was no significant difference between two treatment group.Pathology showed that islet cells of Ge Pi Decoction group destroyed less.Conclusion:Ge Pi Decoction could decrease blood glucose and blood lipids,regulate repair impaired islet cells,improve releasing of insulin,insulin resistance and general conditions of rats with type 2 diabetes mellitus.Ge Pi Decoction could reduce 24h urinary albumin and decrease MCP-1,8-iso-PGF2α,TNF-α,IL-6.Findings confirmed that Ge Pi Decoction is an ideal Chinese drugs preparation treating diabetes and reducing diabetic complication.
